摘要
目的：探讨各种麻醉剂对小鼠晶状体透明度的影响及可能机制。方法：分别用10％水合氯醛（400mg／kg）、2％戊巴比妥钠（40mg／kg）及25％乌拉坦（1000mg／kg）腹腔注射麻醉C57／BL6小鼠，扩瞳后于10、20、30、45、60min裂隙灯观察晶状体浑浊情况，于麻醉60min后断颈处死，H-E染色观察晶状体组织学变化，免疫荧光检测晶状体上皮细胞钙蛋白酶-2及水通道蛋白-1（AQP1）的表达。结果：乌拉坦、水合氯醛麻醉组与戊巴比妥钠麻醉组相比，麻醉持续时间较长，晶状体出现浑浊的时间较早，浑浊程度较重；麻醉后可见晶状体前囊膜增厚，上皮细胞呈增生性变化，上皮细胞钙蛋白酶-2表达增强，AQP1表达减弱。结论：麻醉剂可导致小鼠晶状体突发性浑浊，与上皮细胞上钙蛋白酶-2活性表达增加及AQP1活性表达减少有密切关系。
Objective： To explore the influence of a variety of anesthetics on lens opacification of mice and its possible mechanism. Methods： C57/BL6 mice were anesthetized with intraperitoneal injections of 10% chloral hydrate （ 400 mg/kg ）, 2% sodium pentobarbital （40 mg/kg） and 25% urethane （1 000 mg/kg ） respectively. Lens opacification was examined with a slit lamp at 10 min, 20 min, 30 min, 45 min and 60 min after mydriasis, and then the mice were sacrified by dislocating the neck to death at 60 min after being anesthetized. H-E staining was performed in the lens. Immunofluorescence was used to detect AQP1 and Calpain-2 in the lens epithelial cells. Results： Compared to the group of sodium pentobarbital anesthesia, the group of urethane and chloral hydrate anesthesia had a longer duration of anesthesia, earlier time of the lens opacification and a heavier turbidity. The lens anterior capsules were thickened, the epithelial cells were proliferated, the expression of calpain-2 was enhanced, and the expression of AQP1 in the epithelial cells of the lens was decreased after anesthesia. Conclusion： The anesthesia may induce sudden lens opacification of mice. It may be closely related to the up-regulation of the expression of calpain- 2 activity and the down-regulation of the expression of AQP1 activity.
