丙泊酚对兔晶状体氧化损伤的保护作用探讨 被引量:2

摘要 目的:观察丙泊酚在兔子晶状体氧化损伤中对水通道蛋—l(AQP-1)表达的影响,探讨其对晶状体的保护作用。方法:将正常兔晶状体分为4组:正常对照组、氧化损伤组、维生素C组(100μg/mL)、丙泊酚组(4μg/mL、40μg/mL、100μg/mL)。体外培养24h后,观察晶状体混浊情况,分析晶状体的相对灰度值,免疫组织化学检测AQP-1,分光光度计测定超氧化物歧化酶(SOD)、丙二醛(MDA)的变化。结果:与正常组比较,其他各组晶状体有不同程度混浊;与氧化损伤组和VC组相比,丙泊酚组晶状体相对灰度值增加,AQP-1表达增加,SOD活性增高,MDA的含量降低(P<0.05)。结论:丙泊酚可以通过调节晶状体上皮细胞AQP-1的表达和增强抗氧化能力,来减小晶状体损伤程度。 Objective : To investigate the effeets of propofal on the expression of aquapofin - 1 ( AQP - 1 ) of rabbit' s lens oxidative stress and its protection on lens. Metheds: Rabbit's lenses cultured in vitro were divided into 4 groups: control group, oxidative stress group, groups treated with 100μg/ml vitamin C and with propofol (4μg/ml,40μg/ml, 100μg/ml). After 24 hours of incubation, alteration of opacity was observed by photography and picture analysis. The expression of AQP- 1 was detected by immunohistochemistry with semiquantitative analysis. The content of the superoxide dismutase( SOD) and malonyldialdehyde(MDA) were rm.asured by spectrophotometer. Results: Compared with control group, the transpareney of other lenses were different. In the pmpofol group, the relative gray scale of lenses was increased, the contents of MDA was obviously degraded but SOD and expression of AQP - 1 were higher than the oxidative stress and vitamin C( P 〈 0.05). Conclusion: Propofol can protect lens against oxidative stress by maintaining regulating expression of AQP- 1 and production of SOD, MDA in rabbit' s lens.
