摘要
建立了测定谷物中脱氧雪腐镰刀菌烯醇(DON)含量的酶联免疫吸附测定法(ELISA)和高效液相色谱法(HPLC),利用两种方法对谷物样品进行DON含量测定,并对结果进行了比较.结果表明,ELISA法的检测限为0.043μg/mL,玉米、小麦样品的平均回收率分别为96.8%和97.3%;RSD的平均值分别为8.8%和4.4%;HPLC法的检测限为0.1μg/mL,玉米、小麦样品的平均回收率均为92.7%,RSD的平均值分别为6.8%和3.1%.结果显示,ELISA法相对HPLC法快速简便、回收率高.
The two methods ( HPLC and ELISA) for the determination of DON in grains were established and compared. The limit of detection (LOD) of ELISA was 0.043μg/mL, the average recovery rates of corn and wheat samples were 96.8% and 97.3% respectively, with average RSD 8.8% and 4.4%. The LOD of HPLC was 0.1μg/mL, the average recovery rates of corn and wheat samples were both 92.7% , with average RSD 6.8% and 3.1% respectively. The results showed that the ELISA method had higher recovery and simpler sample preparation.
