摘要
目的 :研究姜黄素在体外对 K5 62 及原代白血病细胞的抑制增殖与诱导分化作用 ,观察其对白血病细胞的细胞周期时相的影响。方法 :细胞生长曲线制作、形态学观察、MTT法、联苯胺反应、流式细胞仪检测。结果 :姜黄素对 K5 62 细胞具有明显的抑制作用 ,其 IC5 0 为 5 .46mg/L ,呈剂量效应关系 ;姜黄素使细胞生长停滞于 S期 ,使细胞在 S期大量堆积 ,而不能进入下一个细胞增殖周期 ;光镜下可见细胞质内出现红色区域 ,联苯胺反应阳性率高于对照组 ( P<0 .0 5 ) ;姜黄素对初发组患者原代白血病细胞抑制率显著大于复发组 ( P<0 .0 5 )。结论 :姜黄素在体外对 K5 62 细胞具有抑制增殖与轻微诱导分化作用 ,并能改变其周期时相 。
Objective:To investigate the effects of curcumin on K 562 cell (human erythrocytic leukemia cells) and primary culture of human leukemia cells and to observe the effect of curcumin on leukemic cell cycle stage in vitro.Methods:Cell growth curve,MTT,morphologic observation,benxedine reaction and flow cytometric detection.Results:Curcumin has significant anti proliferative effect on K 562 cells in a concentration dependent manner,the IC 50 of curcumin on K 562 cells is 5.46 mg/L.Slight differentiation induction is observed from morphologic changes and from benzedine reaction.Curcunin leads to growth arrest at S stage in K 562 ,which is detected by flow cytometry.In addition,the inhibitory effect of curcumin on clinical patients' samples is different,the leukemia cells of original team are more sensitive than those of relapse team ( P <0.05).Conclusion:Curcumin can inhibit the proliferation,slightly induce the differentiation and change the cell cycle stage of K 562 ,inhibit the proliferation of primary culture of human leukemic cells.
