摘要
目的为研究姜黄素对阿霉素在细胞内蓄积的影响。方法观察姜黄素在0、1.56、6.25、12.5μg/ml浓度下,对人口腔上皮癌细胞KB及其多药耐药细胞株KBV200和小鼠肝、肾细胞内阿霉素蓄积的影响,并采用荧光分光光度法对不同浓度姜黄素在各种细胞内的蓄积量进行了测定。结果姜黄素可提高阿霉素在KBV200细胞内的蓄积,降低或不影响阿霉素在KB细胞内的蓄积;不影响阿霉素在肝、肾细胞内的蓄积;姜黄素在各种细胞内随着用量的增加蓄积量增加。结论姜黄素增敏阿霉素抗肿瘤作用在KB与KBV200细胞通过不同机制;姜黄素不增加阿霉素对正常组织的毒性。
To study the effect of curcumin(Cur) on adriamycin(ADR) cellular accumulation by fluorescence spectraphotometry in vitro. We have observed that ADR cellular accumulation in KB and KBv200 cell line, moose's liver and kidney cell when Cur in 0, 1.56, 6.25, 12.5 ug/ml. Meantime, Cur cellular accumulation was measured in the four kind cefls. Resuh:Cur can improve ADR cellular accumulation in KBv200 cell line but reduce in KB cell line. There is no influence in mouse's liver and kidney cell. Cur cellular accumulation is raised with concentration. It is revealed that Cur sensitived antitumor effect of ADR by dif ferent mechanism in KB and KBv200 cell line. Simultaneously, Cur didn't improved toxic in normal tissue of ADR.
