摘要
目的 观察GP对P -糖蛋白 (Pgp)高表达急性粒系白血病 (AML)细胞药物敏感性的影响。方法 采用S -P免疫组化法检测Pgp表达。应用MTT比色分析法测定药物敏感性。结果 16.67% ( 2 / 12 )初治患者和 58.33% ( 7/ 12 )难治或复发AML患者Pgp表达增高 (P <0 .0 5) ;Pgp高表达的AML细胞对阿糖胞苷(AraC)和柔红霉素 (DNR)药物敏感性低于低水平表达者 (P <0 .0 1)。无细胞毒剂量GP显著增强AraC和DNR对Pgp高表达AML细胞增殖的抑制作用 (P <0 .0 1)。结论 AML细胞Pgp高表达和其多药抗性相关 。
Objective:To investigate the effect of gypenosides(GP) on chemosensitivity of acute granulocytic leukemia cells with high level expression of P-glycoprotein. Methods:The P-glycoprotein expression was detected by Streptavidin-Peroxidase(S-P) immunohistochemical method,and in vitro chemosensitivity was measured by MTT colorimetric assay. Results: In primary treated AML patients,the Pgp overexpression rate was 16.67%(2/12), and in refractory and relapsed AML patients,the rate was 58.33%(7/12).The sensitivity of AML cells with Pgp overexpression to cytarabine(AraC) and daunorubicin(DNR) was significantly lower than that of AML cells with Pgp lower level expression(P<0.01). With un-cytotoxic dose, GP might significantly promote the proliferation inhibition effects of AraC and DNR on AML cells with Pgp overexpression (P<0.01). Conclussion:Overexpression of Pgp was correlative with multridrug resistance of AML cells.GP may enhance chemosensitivity of AML cells with Pgp overexpression.
