恒河猴血管内皮细胞移植替代角膜内皮细胞后房水中离子浓度的变化

Ionic concentration changes in aqueous humor of rhesusmonkey after corneal endothelial cells being replaced with vascular endothelial cells by transplantation

摘要 目的探索恒河猴血管内皮细胞移植替代角膜内皮细胞后房水中Na+、K+、Ca2+、Mg2+、P3+、Cl-6种离子浓度的变化。方法将体外培养增殖的恒河猴视网膜脉络血管内皮细胞通过前房注射法移植到超声乳化破坏角膜内皮细胞的恒河猴角膜内表,术前及术后定期抽取房水,通过全自动生化分析仪检测房水中Na+、K+、Ca2+、Mg2+、P3+、Cl-的浓度,并与移植前进行对比分析。结果 Na+:术后3个月时,实验组Na+浓度(164.15±0.49)mmol.L-1略低于空白对照组(167.05±0.49)mmol.L-1,差异有统计学义(P<0.05)。其余组间及组内检验浓度差异均无统计学意义(均为P>0.05)。K+、Ca2+、Cl-:组间及组内检验后K+、Ca2+、Cl-浓度差异均无统计学意义(均为P>0.05)。g2+和P3+:实验组中各观察时间点Mg2+、P3+浓度差异有统计学意义(P<0.05),实验组、实验对照组、空白对照组两两相比不同组别、不同时间差异均有统计学意义(均为P<0.05)。结论恒河猴视网膜脉络膜血管内皮细胞移植到角膜内表面后,对房水中Na+、K+、Ca2+和Cl-离子含量无明显影响,细胞内环境基本稳定;对Mg2+浓度的影响较明显,提示着细胞生理状态的改变过程;对P3+离子浓度有一定影响,尚需进一步的研究和证实。 Objective To explore ionic concentration changes ofNa+,K+,Ca2+,Mg2+,P3+,and C l-in aqueous humor of rhesus monkey after replacing rhesus monkey corneal endothelial cells(RMCECs)w ith vascular endothelial cells(VEC)by transplantation.M ethod s Rhesus monkey vascular endothelial cells were culturedin vitroand transplanted to internal surface of rhesusmonkey corneas through anterior chamber injection,where the intrinsic corneal endothelial cells had been destroyed by ultrasonic emulsification.Aqueous humor from subjects were extracted/collected at regular intervals before and after operation to detect the ionic concentration of Na+,K+,Ca2+,Mg2+,P3+,and C l-by automatic biochem ical analyzer.Then test resultswere compared w ith each other and analyzed statistically.R esu lts Na+:ionic concentration of experimental group(164.15 +0.49)was slightly lower than that of blank control group(167.05+0.49)three months after operation(P0.05).The other differences between and w ithin groupswere statistically insignificant(allP0.05).K+,Ca2+,C l-:A ll differences between and w ithin groups were statistically insignificant(allP0.05).Mg2+,P3+:D ifferences w ithin and between experimental group,control group and blank control group were statistically significant(allP0.05).Conc lu sion Transplantation of VEC to the corneal internal surface had no obvious effect on ionic concentration of Na+,K+,Ca2+,and C l-in aqueous humor,intracellular environmentwas basically stable;While it had an obvious effect on ionic concentration ofMg2+,which indicates the change process of cellular physiological state;Transplantation ofVEC to the corneal internal surface had a certain effect on ionic concentration ofP3+,but it stillneeds further study to be confirmed.
