生血丸对低剂量敌百虫暴露小鼠肝、脾、骨髓细胞及外周血淋巴细胞的影响 被引量:1

摘要 目的探讨生血丸对长期接触低剂量敌百虫水溶液小鼠细胞损伤的影响。方法将150只BalB/C小鼠随机分空白组(生理盐水灌胃40天)、模型1组(敌百虫灌胃20天+生理盐水灌胃20天)、治疗组(敌百虫灌胃20天+生血丸灌胃20天)、模型2组(生理盐水灌胃20天+敌百虫灌胃20天)、预防组(生血丸灌胃20天+敌百虫灌胃20天)。敌百虫溶液剂量为2mg/kg,生血丸剂量为2.5g/kg,每日1次。检测各组小鼠肝、脾细胞中羟自由基、超氧阴离子活性、鸟氨酸脱羧酶(ODC)表达的OD值,计算外周血淋巴细胞、骨髓细胞微核率。结果模型1组、模型2组小鼠肝细胞内羟自由基、超氧阴离子活性、ODC表达、外周血淋巴细胞微核率与空白组相比差异有统计学意义(P<0.05),治疗组与模型1组、预防组与模型2组比较,肝细胞内羟自由基、超氧阴离子活性、ODC表达、外周血淋巴细胞微核率差异有统计学意义(P<0.05)。各组小鼠脾细胞内羟自由基、超氧阴离子活性、ODC表达、骨髓细胞微核率未见明显差异(P>0.05)。结论生血丸能有效预防和改善低剂量敌百虫暴露小鼠肝细胞氧化损伤,降低外周血淋巴细胞的微核率。 Objective To study the effects of Blood Increasing Pill (BIP) on the liver, spleen, bone marrow cells (BMC) and peripheral blood lymphocytes (PBL) in mice chronically exposed to low-dose trichlorfon aqueous solution. Methods Totally 150 mice were randomized into the blank group (40 days of gavage with saline), No. 1 model group (20 days of gavage with trichlorfon+20 days of garage with saline), treatment group (20 days of garage with trichlorfon+20 days of gavage with BIP), No. 2 model group (20 days of gavage with saline+20 days of gavage with trichlorfon) and prevention group (20 days of gavage with BIP+20 days of gavage with triehlorfon). The dose of trichlorfon and BIP was 2 mg/kg and 2.5 g/kg respectively administered once daily. The optical density (OD) values of hydroxyl radical, superoxide anion activity and ornithine decarboxylase (ODC) in the liver cells and spleen celts were detected and the micronucleus rates in PBL and BMC were calculated.Results Comparing with the blank group, the OD values of hydroxyl radical and superoxide anion activity in the liver cells and the micronucleus rates in PBL in two model groups were significantly increased (P〈0.05), but the OD values of ODC were significantly decreased (P〈0.05). Both the differences between the treatment group and No. 1 model group and between the prevention group and No. 2 model group were statistically significant in the OD values of hydroxyl radical, superoxide anion activity and ODC in the liver cells and the micronucleus rate in PBL (P〈0.05). There was no significant difference between groups in the OD values of hydroxyl radical, superoxide anion activity and ODC in the spleen cells and the micronucleus rate in BMC (P〉0.05). Conclusion BIP can effectively prevent and relieve the hepatocytes oxidative damage and reduce the micronucleus rate in PBL in mice exposed to low-dose trichlorfon.
