螺旋藻中亚油酸和γ-亚麻酸含量的气相色谱法同时测定 被引量:7

Simultaneous Determination of Linoleic Acid and γ - Linolenic Acid in Spirulina Platensis(GOM.) Geitl. by Gas Chromatography

摘要 应用气相色谱法同时测定螺旋藻中有效成分亚油酸和γ_亚麻酸的含量。选择以石油醚为溶剂 ,用索氏法提取样品 ,以十七酸为内标物 ,在提取物的正己烷溶液中加入H2SO4-CH3OH(体积比1∶9)甲酯化试剂适量 ,在70℃恒温水浴中甲酯化反应10min ,用气相色谱测定含量。亚油酸甲酯线性方程为Y=2.6x-0.0119 ,r=0.9999 ,加标回收率为108.4 % ;γ_亚麻酸甲酯线性方程为Y=2.416x -0.0279r=0.9997 ,加标回收率为107.8 %。该法灵敏、准确、快速 ,重现性好。 A method for the simultaneous determination of both linoleic acid and γ _ linolenic acid in Spirulina platensis(GOM.)Geitl. is introduced by gas chromatography. Linoleic acid and γ _ linolenic acid were extracted from Spirulina platensis(GOM.) Geitl. by petroleum ether. The extracted linoleic acid and γ _ linolenic acid were esterified to form linoleic acid methyl ester and γ _ linolenic acid methyl ester by H2SO4-CH3OH in hexane solvent at 70 ℃ for 10 min. Both esterified products of linoleic acid and γ _ linolenic acid were simultaneously detected by GC with heptadecanoic acid as an internal standard. The equations of linear regression are Y=2.6 x-0.011 9(r=0.999 9), and Y=2.416 x-0.027 9(r=0.999 7), the recoveries are 108.4% and 107.8% for linoleic acid and γ _ linolenic acid, respectively. The presented method is found to be sensitive and accurate.
