二次回归正交旋转组合设计优化白芷香豆素磷脂复合物的制备工艺 被引量:1

Optimization of Preparation Technology for Coumarin from Angelica dahurica Phospholipid Complex by Quadratic Regression Rotation-orthogonal Combination Design

摘要 目的:优选白芷香豆素磷脂复合物的制备工艺。方法:采用四因素二次回归正交旋转组合设计方法,以复合率为指标,选取投料比、白芷香豆素质量浓度、反应温度、反应时间为考察因素,优化白芷香豆素磷脂复合物的制备工艺。结果:最佳制备工艺为以四氢呋喃为反应溶剂,白芷香豆素与磷脂投料比例1∶3,药物质量浓度2 g.L-1,反应温度50℃,反应时间101 min,复合物结合率达98.58%。结论:该优选的制备工艺稳定可行,为白芷香豆素新制剂的进一步研究提供参考。 Objective: To optimize preparation technology of coumarin from Angelica dahurica phospholipid complex. Method: With recombination rate as index, feeding ratio, the concentration of coumarin from A.dahurica, reaction time and temperature were selected as factors, preparation technology of coumarin from A.dahurica phospholipid complex was optimized by four factors quadratic regression orthogonal rotation combination design method. Result: Optimum preparation technology was as following:with tetrahydrofyran as reaction solvent, proportion of coumarin from A.dahurica to phospholipid 1:3, the concentration of coumarin 2 g·L-1, reaction temperature 50 ℃, reaction time 101 min.Recombination rate of the complex was up to 98.58%. Conclusion: This optimized preparation technology was stable and feasible, it could provide reference for further study on new preparations of coumarin from A.dahurica.
