摘要
目的:利用二次回归正交旋转组合设计对绵萆薢多糖(DSPS)提取工艺进行优化。方法:根据中心组合Box-Benhnken实验设计原理,采用三因素三水平的响应面分析法,依据回归分析确定各工艺条件的影响因子,以DSPS得率为响应值作响应面和等高线图。结果:DSPS的优化工艺参数为:提取次数为2次,液料比为52:1,提取时间为1.8h。在此条件下DSPS得率理论预测值为15.86%,通过验证实验,DSPS得率实测值为15.79%。结论:本文所建立的数学模型有意义(P<0.001),可用来对DSPS的提取工艺进行分析和预测。
Objective: To optimize the extraction technique of Dioscorea septemloba polysaccharides(DSPS) by quadratic regression general rotary unitized design.Methods: The method of response surface analysis with 3 factors and 3 levels was adopted according to the Box-Benhnken center-united experimental design principles based on one-factor tests.Then the factors influencing the technological parameters were determined by means of regression analysis.Response surface and contour were finally graphed with the extraction rate of DSPS(%) as the response value.Results: The optimum extraction conditions of DSPS(%) were concluded as follow: extraction times of 2,liquid-solid ratio of 52:1,extraction time of 1.8 hours.Under these conditions,the yield of DSPS is up to 15.86% and its experimental value is 15.79%.Conclusion: The mathematical model that the present study established was significant(P0.001),and it might be applied to analysis and prediction the extraction technique of DSPS.
