摘要
目的 :探讨不同压力状态对动脉平滑肌增殖的调控作用 ,由此判断冠脉再狭窄过程中压力因素所起的作用。方法 :用含 10 %小牛血清的RPMI- 16 40培养液体外培养新西兰白兔的主动脉平滑肌细胞至 3~ 7代 ,将含 5 %小牛血清的RPMI- 16 40培养液与动脉平滑肌细胞的混悬液 10 0 μl加入 96孔培养板中培养。 2 4h后按照实验分组分别在 6 .6 5kPa ,13.3kPa,19.95kPa ,2 6 .6kPa压力状态下持续培养 48h ,采用四唑盐比色法和3 H -TdR参入法检测平滑肌的生长状况。结果 :13.3kPa压力状态对兔主动脉平滑肌细胞有显著的促增殖作用 (P <0 .0 5 ) ;6 .6 5kPa,19.95kPa ,2 6 .6kPa压力状态对兔主动脉平滑肌细胞无显著的促增殖作用 (P >0 .0 5 )。结论 :生理血压的 1.5~ 2倍压力状态对兔动脉平滑肌细胞增殖有显著的促进作用。
Objective: To investigate the adjusting effect of different pressure state on the proliferation of aortic smooth muscle (ASMCs), and observe if the pressure is one of etiological factors of coronary restenosis after percutaneous transluminal angioplasty (PTCA) and in-stent. Methods: New Zealand rabbit ASMCs were cultured in vitro with RPMI-1640 including 10 percent calf plasma. 100ul mixture of ASMCs and RPMI-1640 including 5 percent calf plasma was put in every cavity of 96-hole plate to culture for test according to the experimental groups, then ASMCs are cultured for 48 hours separately under 6.65Kpa, 13.3Kpa, 19.95Kpa and 26.6Kpa. The numbers of ASMCs proliferation was monitored by MTT method and 3H-TdR incorporation. Results: ASMCs proliferation was significantly enhanced under 13.3Kpa pressure state ( P <0.05), but this phenomenon was not show in 6.65Kpa, 19.95Kpa and 26.6Kpa groups ( P >0.05). Conclusions: 1.5~2 times pressure more than normal blood pressure can significantly enhance the proliferation of ASMCs, this may be one of etiological factors of coronary restenosis after PTCA and in-stent.
