摘要
为探讨MCM3和Ki-67在甲状腺乳头状癌、结节性甲状腺肿及正常甲状腺组织中的表达及意义。采用通过逆转录聚合酶链反应(RT-PCR)检测MCM3在甲状腺乳头状癌、结节性甲状腺肿及正常甲状腺组织中的表达的方法,用免疫组织化学Sp法测定MCM3和Ki-67在甲状腺乳头状癌、结节性甲状腺肿及正常甲状腺组织中的表达。结果显示:(1)DRT-PCR:MCM3与Ki-67在正常甲状腺、癌、结节性甲状腺肿中均有表达。(2)免疫组化:MCM3在正常甲状腺、癌、结节性甲状腺肿组中表达水平有差异(P<0.001),癌和结节性甲状腺肿组与正常甲状腺组表达水平有差异(P<0.01 25);Ki-67在结节性甲状腺肿组和正常甲状腺组中表达尚无差异(P>0.0125),余两两比较表达水平有差异(P<0.0125);MCM3与Ki-67在甲状腺癌组和结节性甲状腺肿组中表达水平有差异(P<0.001)。由此可知,MCM3可作为一种新的甲状腺乳头状癌标记物,较Ki-67更为可靠,从而为判断甲状腺良恶性病变提供一条新的路径。
To explore the significance and expression of MCM3 and Ki-67 in normal thyroid,carcinoma and nodular goiter.RTPCR was employed to examine the level of MCM3 and Ki-67 in the normal thyroid tissue,papillocarcinoma,and nodular goiter. SP immunohistochemical technique was used to observe the expression of MCM3 and Ki-67.Results:(1) RT-PCR:Both MCM3 and Ki-67 were expressed in the normal thyroid tissue,papillocarcinoma,nodular goiter.(2) SP:The expression of MCM3 in the normal ihyroid tissue,thyroid papillocarcinoma and nodular goiter was statistically significant(P0.001).The expression level of IVICiMS in thyroid papillocarcinoma and nodular goiter compared with the expression level in the normal thyroid tissue was statistically significant(P0.0125).The expression of Ki-67 had no significant difference between normal thyroid tissue and nodular goiter(P0.0125),others had significant difference(P0.0125).The expression of MCM3 and Ki-67 had significant difference in carcinoma and nodular goiter(P0.001).MCM3 may be used as a new marker of thyroid carcinoma,more reliable than Ki-67.offering a new approach to differentiate benign and malignant lesions of thyroid.
