摘要
目的:探讨益气化瘀补肾方对兔膝骨关节炎(KOA)模型软骨细胞Ⅱ型前胶原基因(Col2A1)、Ⅹ型胶原基因(ColⅩ)及基质金属蛋白酶-13(MMP-13)mRNA表达的影响。方法:36只新西兰兔随机分为假手术组、模型组和益气化瘀补肾方组,每组12只。模型组和益气化瘀补肾方组家兔采用改良Hulth方法复制KOA模型,假手术组仅切开膝关节表面的皮肤。益气化瘀补肾方组家兔于造模4周后连续用药1个月。观察各组软骨病理形态学变化及Col2A1、ColⅩ和MMP-13 mRNA变化。结果:模型组关节软骨结构破坏较严重,而益气化瘀补肾方组家兔关节软骨退变较模型组明显减轻。益气化瘀补肾方组家兔膝关节软骨Col2A1 mRNA表达明显高于模型组(P<0.01),ColⅩ、MMP-13 mRNA表达明显低于模型组(P<0.05,P<0.01),但与假手术组比较仍有显著性差异(P<0.05,P<0.01)。结论:益气化瘀补肾方可通过促进软骨合成Col2A1和抑制MMP-13及ColⅩ胶原以延缓KOA家兔关节软骨的退变。
Objective： To study the effects of “Yiqi Huayu Bushen Decoction”on expressions of type H collagen （ CoL2A1 ）, type X collagen （ Col X ） and matrix metalloproteinase-13 （ MMP-13 ） mRNA of cartilage cells in knee osteoarthritis （KOA） rabbits. Methods： Thirty-six New Zealand male rabbits were randomly divided into sham group, control group and treatment group with “Yiqi Huayu Bushen Decoction”, with 12 rabbits in each group. The rabbits in the control group and treatment group were modeled to be of knee osteoarthritis by improved Hulth methods ; and the rabbits in the sham group were cut on the knee skin only. The rabbits in the treatment group were treated with “Yiqi Huayu Bushen Decoction” 4 weeks after modeling for one month. The pathological morphology and expressions of CoI2A1, Col X and MMP-13 mRNA of cartilage cells were observed. Results： Histological analysis showed that the structure of articular cartilage was seriously destroyed in the control group, and the degenerative changes of the arlicular cartilage in the treatment group were milder. Compared with the control group, the expression of Cl2A1 mRNA was up-regulated and the expressions of Col X and MMP-13 mRNA were down-regulated in the treatment group （ P 〈 0.05, P 〈 0.01 ） ; there were significant differences in these expressions between treatment group and sham group （P 〈0.05, P 〈0.01 ）. Conclusion： “Yiqi Huayu Bushen Decoction” can promote the synthesis of type 11 eollagen in cartilage cells and suppress the expressions of ColX and MMP-13 mRNA to prevent articular cartilage degradation in KOA rabbits.
