摘要
目的:明确开心散提取物经大鼠灌胃后有效抗抑郁成分的吸收情况。方法:用水蒸气蒸馏法回收石菖蒲挥发油部分,用50%乙醇提取开心散复方有效成分,按原方比例予Wistar大鼠灌胃,分离含药血清和脑组织。含药血清经HPLC检测,定性分析有效抗抑郁成分人参皂苷Rb1、人参皂苷Rg1、3,6,-二芥子酰基蔗糖;脑组织匀浆离心后分离上清液,经萃取后,GC-MS定性分析复方中石菖蒲挥发油有效抗抑郁成分α-细辛醚和β-细辛醚入脑情况。结果:HPLC得到人参皂苷Rb1、人参皂苷Rg1、3,6,-二芥子酰基蔗糖;GC-MS得到α-细辛醚和β-细辛醚。结论:开心散的5种有效抗抑郁成分均能被大鼠吸收,进一步明确了开心散抗抑郁疗效的物质基础。
Objective:To definitely show the absorption of effective antidepressant components after the rat is treated by Kaixin Powder(Radix polygalae,Acorus gramineus soland,Radix et Rhizoma ginseng and Poria).Methods:The Acorus gramineus volatile oil was obtained by vapour distillation and the active constitutions of Kaixin Powder were obtained with ethanol containing 50% water.Wistar rats were orally given the medicine according to the original proportion of Kaixin Powder,the medicinal serum and brain tissue were immediately separated.The medicinal serum was detected with HPLC.The situation of effective antidepressant compositions like Ginsenoside Rb1,Ginsenoside Rg1 and 3,6'-disinapoylsucrose absorbed into blood were analysed qualitatively by qualitative analysis.The brain tissue was made centrifugation to remove debris after grounded into homogenate.The liquid supernatant was extracted to analyse the situation of the effective antidepressant components of the Acorus gramineus volatile oil in the whole formula such as α-Asarone and β-Asarone passed through blood-brain barrier with GC-MS method qualitatively.Results:In the fingerprint of HPLC,three kinds of effective antidepressant components including Ginsenoside Rb1,Ginsenoside Rg1 and 3,6'-disinapoylsucrose were identified.The fingerprint of GC-MS showed that α-Asarone and β-Asarone were identified.Conclusion:The five kinds of effective antidepressant components contained in the whole formula of Kaixin Powder could totally be absorbed by the rats,which further clarifies the material base of antidepressant effect of Kaixin Powder.
