摘要
目的:观察人参皂甙 Rg1对 Aβ_(1-40)诱导大鼠皮层神经元凋亡的影响以及 Rg1对 Caspase-3活性的影响。方法:用吖啶橙-溴化乙锭(AO-EB)染色、TUNEL 染色、DNA 琼脂糖凝胶电泳方法观察神经元凋亡形态学和生态改变;荧光分光光度计法检测凋亡效应分子 Caspase-3活力的变化;RT-PCR 法检测 Cas-pase-3 mRNA 的表达水平。结果:人参皂甙 Rg1预处理可显著降低 Aβ_(1-40)诱导大鼠皮层神经元的凋亡率,凋亡细胞特有的 DNA 梯形条带消失,Caspase-3活力下降,Caspase-3 mRNA 表达下调。结论:人参皂甙 Rg1可通过抑制 Caspase-3的激活,使 Aβ_(1-40)诱导的大鼠皮层神经元减少凋亡。
