摘要
目的:比较远志、石菖蒲中活性成分远志皂苷B、细叶远志皂苷、3,6’-二芥子酰基蔗糖、β-细辛醚对阿尔茨海默病神经干细胞模型的保护、促进增殖分化作用。方法:转染高表达APP基因NSCs (APPNSCs)为模型,检测细胞活力及LDH释放量,测量神经球直径并绘制生长曲线,检测APP-NSCs向星形胶质细胞、神经元、少突胶质细胞分化比率。结果:4种药物均提高APP-NSCs存活率,降低LDH释放量。β-细辛醚、远志皂苷B、3,6’-二芥子酰基蔗糖促进APP-NSCs增殖作用显著,细叶远志皂苷较弱。促进NSCs向神经元分化效果3,6’-二芥子酰基蔗糖最好,β-细辛醚次之,远志皂苷B再次之,细叶远志皂苷没有显著效果。结论:在APP-NSCs保护作用及促进增殖及促进向神经元分化作用中3,6’-二芥子酰基蔗糖效果最好,β-细辛醚次之,远志皂苷B再次之,细叶远志皂苷有细胞保护作用,但没有显著促进NSCs增殖及向神经元分化效果。
Objective: To compare the neuroprotective andproliferation and differentiation promoting effects of 4 active constituents(3,6’-disinapoyl sucrose,tenuifolin,onjisaponin B,β-asarone) from Polygala tenuifolia and Acorus tatarinowii on Alzheimer’s disease neural stem cells(NSCs). Methods: The cell model was constructed by transfection of APP high expression genes into NSCs(APP-NSCs). MTT and LDH assays were used for verifying the neuroprotective effects. The diameters of NSCs were determined and the growth curvewas drawn for verifying the proliferation promotion effects. The ratio of APP-NSCs differentiated to astroglial,neuron,and oligodendrocytes was detected by immunofluorescence cytochemistry. Results: The 4 ingredients could all improve the survival rate of APP-NSCs and decrease the release of LDH. 3,6’-disinapoyl sucrose,onjisaponin B and β-asarone had a significant role in promoting the proliferation and differentiation of APP-NSCsinto neurons. The effect of tenuifolin was relative weak. Conclusion: 3,6’-disinapoyl sucrose hasthe most significant neuroprotective andproliferation and differentiationpromoting effects on APP-NSCs,followed by β-asarone,the third is onjisaponin B,and tenuifolin is relatively weak.
