苦参碱促进三苯氧胺诱导乳腺癌Bcap-37细胞凋亡的机制研究 被引量:5

摘要 [目的]研究三苯氧胺(TAM)联合低剂量苦参碱对人乳腺癌Bcap-37细胞的增殖抑制及作用机制。[方法]用MTT法检测各组细胞的抑制率,克隆形成实验检测对细胞增殖的影响,流式细胞仪检测细胞凋亡率,Western blot法检测p53、Bax、Bcl-2蛋白表达。[结果]TAM对Bcap-37细胞增殖具有明显抑制作用,且具有剂量和时间依赖性,与低剂量苦参碱联合,其抑制作用明显增强。药物作用24、48、72h后,与对照组相比,TAM组的凋亡率均明显增高(P<0.01);与TAM组相比,联合组的凋亡率均明显增高(P<0.01)。Western blot检测发现,与对照组相比,TAM组p53、Bax蛋白表达增加,Bcl-2蛋白表达降低;与TAM组相比,联合组p53、Bax蛋白表达增加,Bcl-2蛋白表达降低。[结论]低剂量苦参碱能促进TAM对Bcap-37细胞增殖抑制作用,其作用机制与增加p53、Bax蛋白表达,降低Bcl-2蛋白表达,促进细胞凋亡有关。
