摘要
采用苦参提取液对TAⅡ小鼠MA737乳腺癌及Hela细胞、λDNA进行了体内外抗癌作用及免疫效应的实验观察。结果表明:1)Hela细胞对苦参碱的药物敏感性高于正常细胞,ID50比值为1.25。2)苦参碱可以明显减弱Hela细胞的电泳迁移速度,减慢率为16.85%。3)苦参碱可与溴化乙锭竞争结合DNA,表明细胞核是药物作用敏感靶位。4)苦参碱改变了DNA的分子构象,从而改变了限制酶对其固有识别顺序的限制性切割。5)荷MA737乳腺癌小鼠经苦参碱治疗TH/TS比值明显提高。6)具生物反应调节作用的苦参碱,不但加强了LAK细胞的免疫效应,尚提高了肿瘤细胞对LAK细胞的敏感性。因此,与苦参碱联合应用,可促进LAK细胞的体内治疗效果。
The effect of AF-7 on MA737 mammary carcinoma ( a trans-plantable breast cancer model )in TAII mice in vivo and HeLa cells in vitro have been studied. It was shown that: (1) the chemotherapeutic sensitivity to AF-7 of HeLa cells was higher than normal cells with a ratio of ID50 being 1. 25; (2) the decreased rate of electrophoretic mobility by AF-7 was 16. 86% ; (3) the binding of AF-7 to DNA competing with EB indicated that the nucleus being the target site of AF-7 action; (4) the molecular structural pattern of DNA must have been altered by adding AF-7 and in turn ,the site of excision by restriction enzyme have been changed as well; (5) the ratio of TH/TS on TAII mice bearing MA737 mammary carcinoma was elevated; (6) anticancer effect of LAK cells was enhanced by AF-7 in vivo and/or in vitro.
