摘要
目的 建立一种能够连续测定小鼠耗氧量动态变化的简易装置和方法。方法 用广口瓶、碱式滴定管、烧杯和连接管道建立小鼠耗氧量测定装置。两组小鼠分别灌胃给予普萘洛尔 2 0mg·kg-1和生理盐水 2 0ml·kg-1,应用该装置观察两组小鼠的缺氧存活时间 ,并测定小鼠耗氧量的动态变化。结果 普萘洛尔组小鼠的缺氧存活时间延长 4 3 35 %(P <0 0 1) ,存活期内的总耗氧量降低 4 3 4 2 % (P <0 0 1) ,其中前 10min的耗氧量降低尤为明显。结论 本测定装置具有简便易行、结果准确、可连续测定小鼠耗氧量动态变化等特点 。
Aim To establish a set of apparatus and method for the continued measurement of the oxygen consumption in mice. Methods The apparatus consists of a bottle with broad-neck for mice, basic burette, beaker containing water and some tubes for connection. Twenty mice were divided randomly into two groups and administered intragestrically propranolol 20 mg·kg -1 or equal volume of normal saline, respectively. The survival time and oxygen consumption were measured in mice subjected to normobaric and hypoxia.Results The survival time was prolonged by 43.35% (P<0.01) and the total oxygen consumption for period of survival time was decreased by 43.42% (P<0.01) in mice received propranolol. Conclusions The apparatus is simple and accurate. And it is suitable for simultaneous observation of survival time and continued measurement for oxygen consumption in mice.
