摘要
目的:观察N-甲基-D-天冬氨酸受体拮抗剂卓西平马来酸盐、艾芬地尔、CGP-37849、L-701.324、HA-966和AMPA受体拮抗剂NBQX分别对培养大鼠海马神经元细胞活力的影响。方法:取孕18天的SD胎鼠海马神经元细胞原代培养,培养的细胞在加入上述药物后,在培养第2天和第7天行台盼蓝染色观察细胞存活情况,四甲基偶氮唑盐法检测细胞存活率,TUNEL法检测细胞凋亡。结果:各浓度的卓西平马来酸盐对培养大鼠海马神经元活力均产生显著毒性。高浓度艾芬地尔和L-701.324可诱导细胞产生凋亡,而低浓度则没有显著作用。各浓度的CGP-37849、NBQX和HA-966对细胞存活无显著影响。结论:只有非选择性NMDA阻断剂MK-801,可降低大鼠海马神经元细胞活力并诱导其凋亡。
Objective:To study the effects of diverses glutamate receptor antagonists including MK-801, ifenprodil, CGP-37849, L-701.324, HA-966 and NBQX on cultured hippocampal neurons vitality. Methods: Hippocampal neurons of pregnant 18d SD fetal rats are in primary culture. Trypan blue staining of cells cultured in the addition of these drugs in 2d and 7d after the culture and MTT assay were used to detect cell viability. TUNEL assay was used to find cells apoptosis. Results'MK-801 of different concentrations showed significant toxicity on hippocampal neuronal development. Ifenprodil and L-701.324 induced cell apoptosis only at high concentrations,while there were no effects on cell viability in each concentration of CGP-37849, HA-966 and NBQX. Conclusion:Only non-selective NMDA blockerMK-801 decreased cell viability and induced apoptosis in neuronal development process.
