产左聚糖枯草芽孢杆菌的突变选育及多糖产物的生物物理分析 被引量:1

摘要 本研究对多糖产生菌株sp.54A-42进行了16SrDNA序列比对分析,鉴定其为枯草芽孢杆菌。对发酵所得多糖产物进行了薄层层析、高效液相色谱、红外光谱和核磁共振分析,证明发酵产物为左聚糖(β-2,6-果聚糖)。通过紫外-LiCl复合诱变方式选育,得到突变株sp.Z-49,多糖产量达到16.1g/L,比出发菌株提高了34.2%,经5次传代发酵实验产量稳定,为进一步研究左聚糖的发酵生产打下了良好基础。 The polysaccharides-producing strain sp.54A-42 was identified to be Bacillus subtilis based on deter-mination and alignment of its 16S rDNA sequence. Its fermenting polysaccharides were proved to be levan (β-2,6-fructan) through the biophysical analysis of thin layer chromatography, high performance liquid chro-matography, infrared spectroscopy and nuclear magnetic resonance. By using the approach of ultraviolet-LiCl compound mutagenesis, the mutant sp.Z-49 was obtained, and the polysaccharides yield of this mutant reached 16.1 g/L with increase of 34.2% compared with the starting strain, which remains a stable yield after five inoculm passages. This research may lay a good foundation for further study on levan producing by fermentation.
