摘要
目的探讨应用purmorphamine特异性激活SHH信号通路对大鼠脑缺血区血管再生的影响。方法采用线栓法制备大鼠急性大脑中动脉脑缺血模型,将大鼠随机分为假手术组、模型组、给药组,其中给药组给予purmorphamine腹腔注射。于术后6h、12h、24h、48h通过免疫组化测定脑缺血后不同时相各组CD105和VEGF表达,PCR测定不同时相各组PTCH-1表达。结果模型组VEGF、CD105、PTCH-1表达明显高于假手术组(P<0.05);给药组与模型组相比较各时间点VEGF、CD105和PTCH-1表达明显增多,差异有统计学意义(P<0.01)。结论激活SHH通路可增加脑缺血组织的VEGF和CD105表达,促进急性脑缺血大鼠的血管再生。
Objective To explore the effect of specific activation of the Sonic hedgehog signaling(SHH) pathway on angiogenesis in rats with cerebral ischemia.Methods The model of acute injury in rats was made by middle cerebral artery occlusion.All the rats were randomly divided into three groups:sham operation group,model group and treatment group,while the treatment group was given intraperitoneal injection of purmorphamine,an specific activator of SHH pathway.At the time point of 6h,12h,24h and 48h after cerebral ischemia,the expression of CD105 and VEGF in rats was observed by immunohistochemistry and the expression of PTCH-1 was detected by RT-PCR.Results The expression of VEGF,CD105 and PTCH-1 of sham group was lower than those of the model group and treatment group.The expression level of VEGF,CD105 and PTCH-1 was higher in treatment group than in model group(P0.05).Conclusion Specific activation of the SHH pathway in acute cerebral ischemia could increase the expression of CD105 and VEGF,which can be useful to promote angiogenesis in rats with cerebral ischemia.
