摘要
目的 探讨诱导型一氧化氮合酶 (i NOS)在急性病毒性心肌炎心肌内的表达及其与心肌损害的关系。方法 6~8周龄 BAL B/ C雄性小鼠 2 4只 ,随机分为两组 :感染组 16只以柯萨奇病毒 B3m(CVB3m)腹腔内注射 ,另 8只为正常对照组。采用免疫组化法检测两组小鼠心肌组织内 i NOS表达 ,并将其半定量与心肌病理变化程度进行相关分析 ,用比色法测定了血清中 NO间接含量。结果 1感染组小鼠心肌标本病理组织学检查均有不同程度的炎性细胞浸润及邻近心肌细胞的变性 /坏死 ;心肌病变积分为 2 .0 8± 1.47。感染组心肌组织内明显表达了 i NOS,i NOS阳性信号密度为 0 .2 44± 0 .0 86 ,而对照组则无阳性发现。2心肌病变积分的变化与 i NOS阳性信号面密度的变化呈显著正相关 (r=0 .85 ,P<0 .0 1)。3感染组小鼠血清中 NO间接含量明显高于对照组 (P<0 .0 1)。结论 CVB3心肌炎小鼠心肌组织内表达了 i NOS,且由此催化产生的 NO在本病的发病中起着重要作用。
Objective To study the expression of inducible nitric oxide synthase (iNOS) in the myocardium of viral myocarditis and the relationship between the expression levels and myocardial damages.Methods Twenty four BALB/C mice of 6 to 8 weeks were randomly divided into two groups:16 were injected intaperineally with CVB 3m and 8 served as control group.Immunohistochemistry technique was used to detect iNOS expression in the myocardium,and myocardial lesions were observed histopathologically.Furthermore,the relationship between iNOS expression levels and myocardlal damages were studied statistically by semi quantitative analysis.In addition,the serum nitrate contents were measured by using fluorospectrophotometry Results Histopathological study on the myocardium from infected mice revealed focal or diffuse lesions characterized by inflammatory cells and degeneration or necrosis of myocytes.The positive staining signals for iNOS proteins which mainly located in the infiltrating inflammatory cells and degenerative or necrotic myocytes were markedly detectable whereas there were no positive findings in the myocardium of control mice.The myocardial lesion score showed significantly positive correlation with the iNOS expression level (P<0 01) .The serum nitrate contents was significantly increased in infected mice as compared with the controls (P<0 01).Conclusion iNOS was expressed in the myocardium of viral myocar ditis murine model induced by CVB 3m .The large amounts of NO produced by iNOS may play an important role in the pathogenesis of viral myocarditis.
