摘要
目的探讨LIGHT在实验性病毒性心肌炎(VM)小鼠中的表达及其意义。方法 60只小鼠随机分为心肌炎组(n=50)和对照组(n=10),心肌炎组腹腔接种0.1 mL柯萨奇B3病毒(CVB3)复制VM动物模型,于接种后7、14和28 d处死,对照组腹腔接种0.1 mL Eagle’s液,于第28天处死,采用RT-PCR、免疫组织化学检测心肌LIGHT mRNA及蛋白表达,ELISA测定血清LIGHT浓度,HE染色检查心肌病理变化,并对心肌炎组血清LIGHT浓度与心肌病变积分进行相关性分析。结果心肌炎组各时点心肌LIGHT mRNA及蛋白表达水平、血清LIGHT浓度均显著高于对照组(P<0.05),并且心肌炎组血清LIGHT浓度与心肌病变积分呈正相关(r=0.81,P<0.05)。结论 LIGHT可能在VM发病机制中起重要作用,血清LIGHT浓度能反映心肌炎的严重程度,可作为VM病情判断的一个血清学指标。
[Objective] To explore the expression and significance of LIGHT in mice with experimental viral myocarditis (VM). [Methods] Sixty mice were divided into two groups randomly. Mice in the VM group (n =50) were inoculated intraperitoneally with 0.1 mL of CVB3. Contrd mice (n =10) were treated with 0.1 mL of Eagle's solution. Ten infected mice were sacrificed on days 7, 14 and 28 after inoculation, and the control mice were killed on day 28 postinoculation. The expression levels of myocardial LIGHT mRNA and protein were examined by RT-PCR and immunohistochemistry. Serum LIGHT concentration was detected using ELISA. Myocardial histopathology was determined with hematoxyiin and eosin stain. In addition, the relationship between serum LIGHT concentration and myocardial histopathological scores was analyzed statistically in VM group. [ Results] The expression levels of myocardial LIGHT mRNA and protein and serum LIGHT concentration from days 7 to 28 after inoculation in VM group were increased significantly compared with control group (P 〈0.05). Serum LIGHT concentration showed significantly positive correlation with the myocardial histopathologic scores in VM group (r=0.81, P〈0.05). [Conclusion] LIGHT may play a pivotal role in the pathogenesis of VM. Serum LIGHT concentration can reflect the severity of myocarditis, and serve as a serumal indicator of pathogenetic condition for VM.
