基于TLR4/NF-κB信号通路探讨丹参酮ⅡA对病毒性心肌炎模型小鼠的心肌保护作用 被引量:1

Myocardial Protective Effect of TanshinoneⅡA on Model Mice with Viral Myocarditis Based on TLR4/NF-κB Signal Pathway

摘要 目的:探讨丹参酮ⅡA通过Toll样受体4(TLR4)/核转录因子κB(NF-κB)信号通路对病毒性心肌炎模型小鼠的心肌保护作用。方法:无特定病原体(SPF)级4~6周龄雄性健康BALB/C小鼠75只,按随机数字表法分为对照组、模型组、丹参酮ⅡA低浓度组、丹参酮ⅡA高浓度组及阳性对照组,每组15只。模型组、丹参酮ⅡA低浓度组、丹参酮ⅡA高浓度组及阳性对照组分别给予100的TCID50病毒液0.1 mL腹腔注射,对照组给予不含病毒的Eagle′s氏液0.1 mL腹腔注射。病毒接种30 min后,模型组和对照组分别给予0.9%NaCl溶液10 mL/kg灌胃,每日1次;丹参酮ⅡA低浓度组、丹参酮ⅡA高浓度组分别给予2.5 mg/mL、5.0 mg/mL丹参酮ⅡA溶液10 mL/kg灌胃,每日1次;阳性对照组给予6.7 mg/mL利巴韦林溶液灌胃,每日1次,连续给药14 d。各组选取13只小鼠,检测比较其心脏指数、血清心肌损伤指标水平、心肌组织病理积分、血清及心肌组织炎性细胞因子水平、心肌组织TLR4、NF-κB表达水平。结果:与模型组比较,丹参酮ⅡA低浓度组、丹参酮ⅡA高浓度组、阳性对照组小鼠心脏指数及血清肌酸激酶同工酶(CK-MB)水平、心肌肌钙蛋白(cTnT)水平、心肌组织病理积分、血清及心肌组织肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)水平及心肌组织TLR4、NF-κB表达水平均降低(P<0.05),且丹参酮ⅡA高浓度组和阳性对照组以上指标均低于丹参酮ⅡA低浓度组(P<0.05)。结论:丹参酮ⅡA能有效减轻病毒性心肌炎小鼠心肌损伤和炎症反应,且呈现一定量效关系,其心肌保护作用可能与抑制TLR4/NF-κB信号通路活性、减少炎性细胞因子释放有关。 Objective:To analysis the myocardial protective effect of tanshinoneⅡA on model mice with viral myocarditis based on Toll like receptor 4(TLR4)/nuclear factorκB(NF-κB)signal pathway.Methods:A total of 75 healthy male BALB/C mice aged 4-6 weeks without specific pathogen(SPF)were divided into control group,model group,tanshinoneⅡA low concentration group,tanshinoneⅡA high concentration group and positive control group according to random number table method,with 15 mice in each group.Viral myocarditis mice models were established by intraperitoneal injection of Coxsackie virus B3 in model group,tanshinoneⅡA low concentration group,tanshinoneⅡA high concentration group and positive control group,and mice in the control group were given intraperitoneal injection of equal volume Eagle′s solution.After 30 min of virus inoculation,the model group and the control group were given 0.9%NaCl solution 10 mL/kg intragastric administration,once a day.TanshinoneⅡA low concentration group and tanshinoneⅡA high concentration group were given 2.5 mg/mL and 5.0 mg/mL tanshinoneⅡA solution 10 mL/kg by intragastric administration respectively,once a day.The positive control group was given 6.7 mg/mL ribavirin solution by gavage once a day.All mice were administered continuously for 14 days.Thirteen mice were selected from each group to detect and compare the cardiac index,serum myocardial injury index,pathological score of myocardial tissue,serum and inflammatory cytokine levels,and expression levels of TLR4 and NF-κB in myocardial tissue.Results:After treatment,compared with model group,the levels of cardiac indexes were lower,levels of serum creatine kinase isoenzyme(CK-MB)and cardiac troponin(cTnT)were lower,the myocardial pathological scores was lower,levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-1β(IL-1β)in serum and myocardial tissue were lower,and the expression levels of TLR4 and NF-κB in myocardial tissue were lower in each group(P<0.05).The above indexes in tanshinone�
