响应面法优化创伤弧菌增菌培养基 被引量:3

Optimization of Vibrio vulnificus Enrichment Medium by Response Surface Analysis

摘要 采用响应面法(response surface method,RSM)对创伤弧菌(Vibrio vulnificus)增菌培养基进行优化。根据因素试验结果,确定蛋白胨、酵母浸膏、NaCl为主要影响因素。在此基础上利用Design Expert 7.1.6软件对RSM法试验结果进行了优化,得到的最优菌培养基组合为:蛋白胨6.0 g/L,牛肉膏1.0 g/L,酵母浸膏4.0 g/L,NaCl 10g/L,FePO4.7H2O 0.01 g/L,pH 7.5。优化后的培养基较3%NaCl碱性蛋白胨水(APW)增菌效果有所提高,并且培养时间缩短。利用优化培养基于35℃恒温摇床中培养10 h,创伤弧菌菌落总数可达2.06×109CFU/mL。 Response surface analysis was used to optimize the enrichment medium for Vibrio vulnificus. According to the prophase single factor experiments, peptones, yeast extract, NaC1 were the significant influence factors on Fibrio vulnificus production. On the basis of this, Design Expert 7.1.6 was used to optimize the results of RSM. The optimized medium comprised of peptone 6 g/L, beef extract 1.0 g/L, yeast extract 4 g/L, NaCl 10 g/L, FePO4.7H2O 0.01 g/L, pH 7.5. The enriching effect of optimization medium was much better than APW, and the culture time was shorter. The total number of colonies of Fibrio vulnificus could reach 2.06×109 CFU/mL in the optimal growth medium at 35 ℃ for 18 h in the constant temperature shaker.
