摘要
[目的]优化超声波辅助双水相系统提取虫草多糖的工艺。[方法]采用正交试验设计对虫草多糖进行双水相提取工艺研究,浓硫酸-苯酚法测定多糖的含量。[结果]虫草多糖的最佳提取工艺为:北冬虫夏草子实体粉末(过100目筛)在60℃,料液比0.03 g/ml,浸提3 h后,超声波辅助提取30 min,虫草多糖的最大得率达到30.1%。[结论]优化出一条全新的虫草多糖最佳提取工艺过程,为虫草多糖的进一步开发奠定了良好基础。
[Objective] To optimize the extraction process of polysaccharide from cultured Cordyceps militaris by ultrasonic assisted aqueous two phase system.[Method] Orthogonal design was used to optimize extracting parameter and the content of polysaccharide was determined with phenol-sulfuric acid.[Result] The optimum extraction conditions of polysaccharide were: fruiting body of powder(100 mesh),solid-liquid ratio of 0.03 g/ml,extraction time of 3 h,temperature of 60 ℃,ultrasonic time of 30 min at which the highest extraction rate of polysaccharides was 30.1%.[Conclusion] The optimum extraction process of polysaccharide was obtained,and provided the basis for its further development.
