Expresssion of BAG-1 in spinal cord after experimental injury

摘要 目的观察Bcl-2结合抗凋亡基因1(BAG-1)在大鼠损伤脊髓中的表达。方法 42只SD大鼠随机分为损伤组(A组,36只)和假手术组(B组,6只)。制作脊髓损伤模型,用Westernblot、免疫组织化学和免疫荧光技术检测BAG-1在损伤脊髓中的表达、组织分布与细胞定位。结果BAG-1在脊髓损伤后8h开始表达增加,2d达到最高峰值,损伤后2周恢复到损伤之前水平。与B组相比,在脊髓损伤后2d,BAG-1的阳性信号在临近损伤中心的脊髓中明显增加。BAG-1与神经元、星形胶质细胞存在共定位,并且BAG-1在这两种细胞中表达上调。结论脊髓损伤后,脊髓中BAG-1的表达上调可能参与神经元的凋亡以及星形胶质细胞活化的病理生理过程。 Objective To observe the expression of Bcl-2 associated athanogene-1(BAG-1) in rat models with spinal cord injury. Methods Fourty-two SD rats were divided into two groups of A (subjected to spinal cord injury, 36 rats) and B(sharrn operated, 6 rats). The expression, patterns and distribution of BAG-1 in the spinal cord were detected by Western blot, immunohistochemistry and immunofluorescence, respectively. Results BAG-1 protein expression was significantly upregulated at 2 hours after injury,which peaked in 2 days and recovered to the baseline in 2 weeks later. Compared to group B, the immunochemical staining in group A was increased significantly in 2 days after injury. BAG-1 expression was collocated and upregulated in the neurons and astrocytes. Conclusion The upregulation of BAG-1 in the injured spinal cord may be associated with the apoptosis of neurons and proliferation of glial astrocytes.
