摘要
目的探讨Bcl-2家族成员Bcl-xl、Bcl-2及Bax在类风湿关节炎(RA)发病过程中的变化,从而进一步明确RA的发病机制。方法 16只雄性Wistar大鼠适应性饲养1周后随机分为正常对照组及胶原诱导的RA模型组,每组8只。在初次免疫后6周处死大鼠,应用免疫组化、RT-PCR及免疫印迹(Western blotting)法检测Bcl-xl、Bcl-2、Bax基因及蛋白质在RA模型组及正常对照组大鼠滑膜组织中的表达情况。结果 RA模型组大鼠滑膜组织Bcl-xl、Bcl-2 mRNA表达水平显著高于正常对照组(P<0.01);正常对照组大鼠滑膜组织可见微量的Bax mRNA阳性表达,但其表达水平仍显著低于RA模型组(P<0.05)。同时,在正常对照组大鼠滑膜组织Bcl-xl、Bcl-2及Bax蛋白质均有少量表达,但在RA模型组三者的表达水平发生了显著变化,其中Bcl-xl、Bcl-2表达水平显著增高(P<0.01),而Bax蛋白表达水平虽比RA模型组略有升高,但差异无统计学意义(P>0.05)。结论在RA发病过程中抗凋亡因子Bcl-xl、Bcl-2呈明显高表达状态,而促凋亡因子Bax的基因及蛋白质水平升高幅度远远低于抗凋亡基因Bcl-xl及Bcl-2升高的水平,造成滑膜细胞增殖/凋亡失衡,促进了RA的发生发展。
Objective To explore the changes of the expressions of apoptosis - related factors Bel - xl, Bel - 2 and Ban in the synovial tissues of rats with rheumatoid arthritis (RA) in order to further understand the pathogcnesis of RA. Methods 16 male Wistar rats were randomly divided into control group and RA model group induced by collagen after one week of adapta- bi!ity breeding, with each group 8 rats. The rats were killed six weeks after primary immune. Immunihistochemistry, RT - PCR and ~;estern blotting were used to detect the changes of the expressions of apoptosis - related factors Bcl - xl, Bel - 2 and Bax in sy^ovia! tissues of RA model group and control group. Results The expression levels of Bcl - xl and Bcl - 2 mRNA in synovial tissue3 of RA model group were significantly higher than those of the control group (P 〈0. O1 ) . A tiny of positive expression of Bax RNN g could be detected in synovial tissues of the control group, but the expression level was still significantly lower than that of the RA model group (P 〈 0. 05 ) . Meanwhile, Bcl - xl, Bcl - 2 and Bax protein all had a small amount of expressions in the synovial tissues of control group, but the expression levels of these three factors were much more significant in RA model group, in which the expression levels of Bcl - xl and Bcl - 2 were significantly increased ( P 〈 O. 01 ), while the expression level ot Bax were higher than that of the model group, but showed no statistically significant difference ( P 〉 O. 05 ) . Conclusion In the pathagenesis of RA, there is 'abnormally high expression of apoptosis inhibitors Bcl - xl and Bc1-2. While the increase of ex- pression levels of gene and protein of apoptosis promoter Bax are significantly lower than the increase of apoptosis inhibitor Bcl - xl and Bcl -2, resulting in the imbalance of proliferation/apoptosis in synovial tissues and the occurrence and development of RA.
