摘要
目的探讨角果木提取物Tagalsin的抗肿瘤作用及其对bcl-2/bax表达的影响,了解其抗肿瘤机制。方法建立小鼠艾氏腹水瘤实体瘤模型,随机分为5组,Tagalsin高剂量组给予Tagalsin 16.50mg/(kg.d)、中剂量组给予Tagalsin 3.30mg/(kg.d)、低剂量组给予Tagalsin 0.66mg/(kg.d),空白对照组给予等体积食用油,阳性对照组给予卡莫氟100mg/(kg.d),连续灌胃给药10d,然后分别计算各组抑瘤率、脾指数和胸腺指数,免疫组化方法检测bcl-2、bax的表达水平及其比值。结果与空白对照组比较,阳性对照组及Tagalsin各剂量组肿瘤生长缓慢(F=1 588.82,t=36.38~75.19,P<0.05),高、中、低剂量组及阳性对照组的抑瘤率分别为37.25%、34.10%、29.51%、61.03%;与空白对照组比较,阳性对照组及Tagalsin各剂量组脾指数和胸腺指数均显著降低;与阳性对照组比较,Tagalsin各剂量组胸腺指数均降低(F=95 874.12,t=2.54~474.59,P<0.05)。阳性对照组及Tagalsin各剂量组bcl-2/bax比值与空白对照组比较差异均具有显著性(F=342.89,t=23.30~47.19,P<0.05)。结论 Tagalsin可能通过下调bcl-2、上调bax的表达,从而抑制肿瘤的生长,且对小鼠的体液免疫有一定的调节作用。
Objective To explore the antitumor action of Tagalsin(extracted from Ceriops tagal) and its effect on the expression of bcl-2/bax. Methods Models of Ehrlich ascites tumor were created in rats,which were randomly divided into five groups as follows: high-dose Tagalsin(Tagalsin 16.50 mg·kg-1·d-1),moderate-dose Tagalsin(3.30 mg·kg-1·d-1),low-dose(Tagalsin 0.66 mg·kg-1·d-1),blank-control and positive control.In Tagalsin groups,a corresponding dose of Talgasin was offered;in the blank group,a same volume of edible oil was given;in positive control group,carmofur tablets 100 mg/(kg·d),which were all administrated intragastrically for 10 days.The oncostatic rate,the spleen and thymus indexes were calculated,and the expressions of bcl-2/bax detected by immunohistochemistry. Results Compared with the blank control group,the tumor grew more slowly in the positive control and different-dose Tagalsin groups(F=1 588.82,t=36.38-75.19,P〈0.05),and the oncostatic rate of the high-,middle-,low-dose and positive control group was 37.25%,34.10%,29.51%,and 61.03%,respectively.Compared with the blank control group,the indexes of spleen and thymus in the positive control and Tagalsin groups were significantly lower;compared with the positive control,the index of thymus in Tagalsin groups was lower(F=95 874.12,t=2.54-474.59,P〈0.05).The differences of the ratio of bcl-2/bax among the blank control,the positive control and Tagalsin groups were statistically significant(F=342.89,t=23.30-47.19,P〈0.05). Conclusion Tagalsin inhibits the growth of tumor by decreasing bcl-2 and increasing bax expression,which has a certain regulation action on humoral immunity in mice.
