摘要
目的采用超滤法研究羌活提取物中异欧前胡素与人血清白蛋白(HSA)的结合作用。方法采用超滤法和高效液相色谱-荧光法测定游离异欧前胡素的质量浓度,计算蛋白结合率,并根据Scatchard作图法计算结合常数。结果当质量浓度为3.2,6.4,13.6,27.7,57.1μmol/L时,异欧前胡素平均HSA结合率分别为56.2%,54.9%,54.4%,49.7%,44.1%。结合常数为2.12×104L/mol,结合位点数n为0.4。结论所用法简单、快速,适合于异欧前胡素蛋白结合率研究。异欧前胡素与人血浆白蛋白具有中等强度的结合。
Objective To study the affinity of isoimperatorin extracted from notopterygium to human serum albumin（HSA） by ultra.filtration method. Methods The uhraflhration method combined with the HPLC-fluorescence method was employed to determine the concentration of free isoimperatorini and calculate the protein binding rate of isoimperatorin and the binding constant according to the Scatehard plot. Results In the plasma concentrations of 3.2, 6. 4i 1,3.6, 27.7, 57. 1 μmol/L, the average HSA binding rates of isoimperatorin were 56.2%, 54.9%, 54.4%, 49.7% and 44.1% respectively. The binding constant was 2.12 × 104 L/mol, and the numbers of binding site was 0. 4. Conclusion The used method is simple, rapid and suitable for the study of protein binding rate of isoimpera- torin. Isoimperatorin possesses the moderate- intensity affinity with HSA.
