50℃热疗对CT-26细胞体外作用的观察 被引量:1

摘要 目的:观察50℃时不同热疗时间对CT-26细胞的损伤作用。方法:将CT-26细胞在50℃水浴中加热至各时间点(5、10、15、20和30min),后采用不同的方法分别观察不同时间热疗后的细胞形态、细胞生存率和细胞生长周期及凋亡情况。结果:细胞生存率由5min时的72.3%降至30min时的1.7%(MTT法),由72.9%降至1.4%(台盼蓝染色排除法)。5min时细胞凋亡率为(28.047±0.675)%,在各时间组中最高,P<0.05。结论:50℃热疗,<5min以诱导细胞凋亡为主;10min时不仅引起细胞坏死,还可诱导细胞凋亡;>15min以细胞坏死为主。 OBJECTIVE: To observe the injury function of thermotherapy to CT-26 cells at the different heating times at the temperature of 50 ℃. METHODS:CT-26 cells were heated in the 50℃ water bath case for 5,10,15,20 and 30 min separately. After the thermotherapy different approaches were taken to observe the cells' shape, the survival rates,the apoptosis and cell cycle of tumor cells. RESULTS:The survival rates decreased from 72.3% to 1.7% (by MTT method)and from 72.9% to 1.4% (by Trypan blue method). When the heating time was 5 min, the apoptosis rate was (28. 047±0. 675)%. It was the highest in all the groups (P〈0.05). CONCLUSIONS:At 50 ℃ ,when the heating time is within 5 min,the apoptosis is mainly shown. When the heating time is 10 min the thermotherapy can not only cause cell death but also induce apoptosis, and when the heating time is beyond 15 minutes cell death is dominant.
