康复新液对小鼠噁唑酮结肠炎的疗效及其机制初探 被引量:9

摘要 背景:我国溃疡性结肠炎的发病率不断升高,现有治疗尚存在不足,需寻找新的治疗方法和药物。目的:观察康复新液对小鼠(?)唑酮(OXZ)结肠炎的疗效,初步探讨其治疗机制。方法:将60只小鼠随机分为空白对照组、模型组、康复新液组、高浓度康复新液组、激素组,以OXZ灌肠诱导结肠炎模型。分别于灌肠后第3、8、13 d处死小鼠。行疾病活动指数(DAI)、结肠大体评分和组织学评分,以ELISA法检测结肠组织髓过氧化物酶(MPO)、白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)、NF-κB含量。结果:灌肠后第3、13 d,与空白对照组相比,模型组DAI、结肠大体评分、组织学评分、MPO、IL-4、TNF-α、NF-κB含量均明显升高(P<0.05),IFN-γ含量无明显差异。灌肠后第8、13 d,给予康复新液和高浓度康复新液治疗后,上述指标均明显改善(P<0.05),IFN-γ含量无明显差异。灌肠后第8 d,激素组仅大体评分和IL-4含量明显降低(P<0.05)。结论:康复新液灌肠可使OXZ诱导的小鼠结肠炎症明显减轻,其作用可能与抑制肠道异常炎症反应、调节细胞因子水平、抑制NF-κB转录有关。 Background: hmidence of ulcerative colitis is increasing in China, available treatment still has some defects and it is necessary to search new treatment and medicine. Aims: To evaluate the therapeutic effect and mechanism of Kangfuxinye on oxazolone (OXZ) induced colitis in mice. Methods: Sixty mice were randomly divided into blank control group, model group, Kangfuxinye group, high concentration Kangfuxinye group and corticosteroid group. Colitis model was induced by OXZ enema. Mice were sacrificed on the 3rd, 8th, 13th day after enema, respectively. Disease activity index (DAI), colonic macroscopic score and histological score were evaluated. Concentrations of myeloperoxidase ( MPO), interleukin (IL) -4, interferon (IFN) -y, tumor necrosis factor (TNF) -ct, NF-KB in colon tissue were determined by ELISA. Results : On the 3rd, 13'h day after enema, DAI, macroscopic score, histological score, and concentrations of MPO, IL4, TNF-c~, NF-KB were obviously increased in model group when compared with blank control group (P 〈 0.05 ) ; no significant difference in concentration of IFN-y was found between the two groups. On the 8'h, 13th day after enema, all the above- mentioned indices were significantly ameliorated in Kangfuxinye and high concentration Kangfuxinye groups ( P 〈 0.05 ) ; the difference in concentration of IFN- was not statistically significant. On the 8'h day after enema, macroscopic score and concentration of IL-4 were significantly decreased in corticosteroid group ( P 〈 0.05 ). Conclusions: Kangfuxinye can ameliorate inflammation in OXZ-induced murine colitis. The therapeutic effect of Kangfuxinye may be related to the inhibiting of abnormal inflammation reaction, modulating level of cytokines, and inhibiting of transcription of NF-KB.
