温燥诱发咳嗽机理的实验研究 被引量:2

摘要 目的通过观察温燥对BALB/c小鼠气道AQP5和Muc5ac表达的影响,探讨温燥诱发咳嗽的机理。方法 SPF级BALB/c小鼠80只,雌雄各半,随机分为4组:正常对照组(C组)、温燥模型Ⅰ组(WⅠ组)、温燥模型Ⅱ组(WⅡ组)、桑杏汤治疗组(S组),置人工气候箱内按"温度-相对湿度-风速"条件进行造模。用实时定量荧光PCR法检测AQP5 mRNA和Muc5ac mRNA在小鼠气道内的表达,用蛋白免疫印迹法和免疫组化法分别检测AQP5和Muc5ac的蛋白表达。结果与C组比较,WⅠ组小鼠气道AQP5 mRNA及其蛋白表达明显减少(P﹤0.01),Muc5ac mRNA及其蛋白表达明显升高(P﹤0.01);这种变化趋势随着温燥作用时间延长而增强,但这种增加的趋势可被桑杏汤抑制。结论温燥可能通过改变AQP5和Muc5ac的表达而诱发咳嗽。 Objective To study the mechanism of cough induced by warm - dryness and to observe the effect of warm - dryness on AQP5 and mucSac expression in airway of BALB/c mice. Methods 80 specific pathogen Free BALB/c mice ,male and female in half, were randomly divided into 4 groups:normal control group (C group), Warm- dryness model I (W I group), Warm -dryness model II (W II group) and Sangxingtang treatment group (S group) ,for modeling in "temperature - relative humidity - the wind" conditions. The expression of AQP5 mRNA and mucSac mRNA in airway was detected by real time quantitative fluorescence PCR ,and the expression of AQP5 and muc5ac protein was assayed by Western Blot and immunohistochemical method, respectively. Results Compared with C group, the expression of AQP5 mRNA and protein in W I was group decreased remarkably ( P 〈 0.01 ), and the expression of mucSac mRNA and protein in W [ group increased significantly ( P 〈 0.01 ). This tendency of change became more obvious as time went by, and it could be restrained by Sangxingtang. Conclusion Cough, induced by warm- dryness,may be due to changes of AQP5 and muc5ac expression.
