摘要
目的通过建立免疫性血小板减少性紫癜(ITP)小鼠动物模型,观察羊蹄根对模型小鼠外周血细胞因子的影响,探讨羊蹄根的免疫调节机制。方法:50只Balb/c小鼠随机分为阴性对照组、模型组、阳性对照组、高剂量和低剂量羊蹄根组。除阴性对照组外,其余各组隔日注射豚鼠抗小鼠血小板血清(GP-APS)建立ITP小鼠模型。注射GP-APS 24h后,阴性对照组和模型组灌服0.9%氯化钠溶液,阳性对照组醋酸泼尼松按5mg/kg灌胃,羊蹄根高剂量和低剂量组按其5、0.5g/kg灌胃。连续给药14d后处死小鼠检测脾脏和胸腺指数;分离血清检测干扰素-γ(IFN-γ)和白细胞介素-4(IL-4)水平。结果:与阴性对照组比较,模型组脾脏、胸腺系数和IFN-γ水平显著升高(P<0.05);IL-4水平显著下降(P<0.05),IFN-γ/IL-4比值升高。与模型组比较,高剂量羊蹄根组脾脏、胸腺系数和IFN-γ水显著下降(P<0.05);IL-4水平显著升高(P<0.05),IFN-γ/IL-4比值降低具有显著差异。结论:高剂量羊蹄根可通过影响Th1/Th2细胞及因子水平而对ITP免疫功能起到调节作用。
Objective:To observe the effect of Rumex.japonicus Houtt extract on peripheral blood cytokines in immune thrombocytopenic purpura mouse model and discuss it's immune regulatory mechanism.Methods:50 Balb/c mice were randomly divided into negative control group,positive control group,model group,high and low dose of Rumex.japonicus Houtt extract groups.ITP mouse model were made by injected intraperitoneally guinea pig-antimouse platelet serum(GP-APS) every other day.Twenty four hours after injecting the GP-APS,the mice in the negative control and model groups were fed with normal saline,those in the positive control group were given with prednisone acetate(5mg/kg),and those in the high(5g/kg) and low(0.5g/kg) dose of Rumex.japonicus Houtt extract groups were administrated with the extract,successively given for 14 days.The coefficients of spleen and thymus were calculated,and levels of IFN-γand IL-4 were detected.Results:The coefficients of spleen and thymus and levels of IFN-γin model group were significantly increased compared with the control group(P〈0.05),and the levels of IL-4 decreased significantly(P〈0.05),thus the ration of IFN-γ/IL-4 increased significantly.The coefficients of spleen and thymus and levels of IFN-γin high dose of Rumex.japonicus Houtt extract groups were significantly decreased compared with the model group(P〈0.05),and the levels of IL-4 increased significantly(P〈0.05),thus the ration of IFN-γ/IL-4 decreased significantly.Conclusion:High dose of Rumex.japonicus Houtt extract can regulate the immune function of ITP by influencing the level of Th1/Th2 cells and their cytokines.
