摘要
目的探讨一种快速同时测定山豆根药材中氧化苦参碱和苦参碱含量的方法。方法采用超高效液相色谱法,色谱柱为WatersACQUITY uplcHSS T3柱(50 mm×2.1 mm,1.8μm),流速0.5 ml/min,进样量5μl;以水-0.1%的三乙胺乙腈溶液为流动相,梯度洗脱;检测波长220 nm。结果氧化苦参碱在20.16~201.6μg/ml、苦参碱在20.88~208.8μg/ml范围内,浓度与峰面积线性关系良好,相关系数(r)均为0.999 9;2种成分的平均回收率分别为99.1%、99.5%,RSD分别为1.27%、1.40%(n=9)。结论 UPLC法快速、准确,可用于山豆根药材的质量控制。特别适合于成分较复杂的中药材的快速分析。
Objective To study the rapid determination of oxymatrine (OM) and matrine (M) in Radix Sophorae Tonkinensis at the same time. Methods A UPLC method was established. The Waters ACQUITY upleHSS T3 column was used. The mobile phase consisted of water-acetonitrile (with 0.1% triethylamine) with gradient elution at the flow rate of 0.5 ml/min ,the detection wavelength was set at 220 nm. Results The linear range of oxymatrine was 20.16-201.6 μg/ml and matrine was 20.88 - 208.8 μg/ml, respectively. The average recoveries; of oxymatrine and matrine were 99.1% with RSD = 1.27%, and 99.5% with RSD = 1.40% (n =9), respectively. Conclusion The method is rapid and accurate, and can be used for the quality control of Radix Sophorae Tonkinensis. The UPLC method is suitable for the rapid detection of Chinese herbal medicines.
