摘要
目的:研究四君子汤及其纳米制剂对抗生素脱污染小鼠肠屏障的影响。方法 :应用林可霉素灌胃制备小鼠肠屏障损伤模型,然后应用常态四君子汤(水提液,15g/kg)及纳米四君子汤(3g/kg)进行治疗,同时设正常、阳性对照丽珠肠乐及自然恢复组,于给药7天后处死小鼠,进行肠道菌群定量分析、同时检测肠粘膜sIgA及肠组织DAO含量。结果:林可霉素灌胃3天后,小鼠肠道菌群失调,肠粘膜sIgA和肠组织DAO含量均较正常对照组减少(P<0.01),持续7天治疗后,和自然恢复组比较肠道双歧杆菌和乳酸杆菌菌量明显上升,肠粘膜sIgA及肠组织DAO的含量明显升高。各治疗组间比较纳米四君子汤的效果要好于常态四君子汤和丽珠肠乐。结论 :四君子汤纳米化后能扶植肠道正常菌群生长,修复损伤的肠粘膜,增强肠粘膜免疫功能,且效果优于常态中药。
To explore the influence of Sijunzi Decoction and its nano-preparation on the intestinal barrier of contaminated-freed mice by antibiotic. Methods: Intestinal barrier injured model mice were established by intragastrie administration of lineomycin and then treated with conventional Sijunzi Decoction(15g/kg) and nano-Sijunzi Decoction(3g/kg). In the same time, group of normal control, group of positive control and spontaneous recovery group were set. After treated for 7 days, the mice were suicide and intestinal flora was analyzed quantitative- ly and the level of slgA of intestinal mueosa and DAO of intestinal tissue were detected. Results : After intragastrie administration of lincomy- sin 3 days, intestinal flora of the mice altered. Compared with normal control, the level of slgA of intestinal mueosa and DAO of intestinal tis- sue decreased ( P 〈 0.01 ). After being treated 7 days, compared with natural recovery group, the quantity of intestinal Bacillus bifidus and lactobacilli of the mice increased differently ( P 〈 0.01 ), and the level of slgA and DAO step up also ( P 〈 0.01 ). The effect of nano-Sijunzi Decoction is better than that of conventional Sijunzi Decoction and Bifidobiogen-Livzon. Conclusion : Nano-Sijunzi Decoction can foster intes- tinal normal flora, repair injured intestinal mueosa and improve intestinal mueosa immune function and have an advantage over conventional Chinese medicine with less dose.
