摘要
为了提高米荞种子的萌发率和建立其愈伤组织培养体系,研究了不同处理方式对米荞种子萌发率的影响以及选择不同激素配比对米荞植物的子叶、胚轴和胚根外植体进行愈伤组织诱导。结果表明,采用去种皮的米荞种子经浸种处理,其种子在24和48 h的萌发率均最高为100%;诱导米荞胚根、胚轴和子叶外植体的最佳培养基配方为MS+2,4-D 1 mg.L-1+6-BA 2 mg.L-1,其中胚轴外植体的愈伤组织诱导率最高为100.00%,子叶为78.90%,胚根为54.44%。
To improve the seed germination rate of FagoFyrwn tatarlcum Gacrtn. and establish the culture system of callus,the different treat- ment methods were used to investigate the seed germination rate of FagoF),rum tataricum Gaertn. The optimum auxin ingredient was screened for callus induction in cotyledon, hypocotyl, radicle explants of Fagoy tatadcum Gaertn. The results showed that,when the FagoFyrum tear/cum Gaertn. seed with no testa was soaked for 24 and 48 h,the germination rate was up to 100 % . The optimum auxin ingredient for callus induction was MS + 2,4-D 1 mg. L-1 +6-BA 2 mg. L-l. When the hypocotyl, cotyledon and radicle were taken as explants,the callus induction rate was up to 100 %, 78.90 % and 54.44 %.
