摘要
目的观察50℃时不同热疗时间对体外培养各种肿瘤细胞热休克蛋白(heat shock protein,HSP)表达的影响。方法①选择4种不同的肿瘤细胞株:慢性髓系白血病敏感型细胞株(K562/S细胞)、人胃癌AGS细胞株(AGS细胞)、BALB/c小鼠来源的结肠癌细胞CT-26株(CT-26细胞)、乳腺癌药物敏感细胞株MCF(MCF细胞)。②选取对数生长期的各肿瘤细胞制成细胞爬片,将细胞爬片分为实验组和对照组。③实验组:将爬片水浴加热各时间段(5分钟、10分钟、20分钟、30分钟、50分钟),后置于37℃、5%CO2的培养箱中继续培养24小时。对照组:将各细胞爬片37℃水浴放置各时间段。免疫组织化学法(immuno-histochemistry,IH)检测热疗后各肿瘤细胞HSP70的表达情况。结果 IH发现,热疗后HSP70的表达量明显提高,在加热到20分钟时各肿瘤细胞中HSP-70强阳性表达,但在加热30分钟后随着细胞大量坏死,HSP-70阳性表达逐渐减弱,在50分钟时HSP-70阳性表达基本消失。结论 HSP70的阳性表达率随时间的延长而提高,直至细胞死亡而停止表达。
Objective: To observe the effects of thermotherapy at the temperature of 50℃ and in different heating times on the expression of HSP70 of tumor cells in vitro.Methods: ① The four kinds of different tumor cells,K562/S cells,AGS cells,CT-26 cells,MCF cells were chosen.② All kinds of tumor cells in logarithmic phase were made into cells’ creep slides and these cells’ creep slides were selected to be divided into two group as the experimental group and the control group.③ In the experimental group cells’ creep slides were heated in the 50℃ water bath case for 5 minutes,10 minutes,20 minutes,30 minutes and 50 minutes separately,while in the control group the same processes were followed only when the heating temperature was 37℃,afterward the cells continued to be cultured normally in the incubator for another 24 hours under 37℃,and 5%CO2.The HSP70 expression of tumor cells was detected by the methods of immunohistochemistry.Results: The results of immunohistochemistry showed that the thermotherapy enabled tumor cell to show positive expression of HSP70,more obvious results were detected under 50℃ for 20 minutes,but in 30 minutes or more-heating-group the HSP70 positive expression decreased gradually because tumor cells were too damaged by thermotherapy,and the HSP70 expression quantity almost disappeared under 50℃ for 50 minutes.Conclusion:The HSP70 expression quantity increases when the heating time prolonges until tumor cells are too damaged by thermotherapy to express the proteins.
