摘要
目的比较银翘散煎剂与5种相关成药(银翘解毒颗粒、银翘解毒片、银翘解毒丸、银翘解毒液、银翘解毒合剂等)的HPLC特征图谱、日服用生药及指标成分量的差异。方法采用HPLC色谱法,phenomenexluna色谱柱(4.6 mm×250 mm,5μm),甲醇-0.1%磷酸溶液为流动相进行梯度洗脱,检测波长为230 nm,流速为1.0 mL/min。结果得到分离度较好的银翘散煎剂HPLC特征图谱,识别了其中的16个特征峰,上市银翘解毒成药如颗粒剂、片剂、丸剂、口服液、合剂分别保留了煎剂中6和10~13的特征峰,绿原酸、连翘酯苷A、牛蒡苷等指标成分日服用量与煎剂相比均有不同程度的降低。结论银翘散煎剂与5种银翘解毒成药HPLC特征图谱、日服用生药及指标成分量存在较大差异,提示基于临床汤剂研发与生产的成药制剂应关注并克服汤剂成药化缺陷。
Objective To investigate the differences in HPLC specific chromatogram, and daily dose of the effective compositions between the decoction of Yinqiao Powder and five Yinqiao Jiedu preparations including granule, tablets, pills, oral liquid, and mixture. Methods High performance liquid chromatography method was developed. The sepa- ration was performed on a phenomenex luna column(4.6 mm × 250 nm, 5 μm)by gradient elution with methanol and 0.1% phosphoric acid as the mobile phase and flow-rate of 1.0 mL· min^-1, and the wave lengths was set 230 nm. Results The HPLC specific chromatogram of the decoction of Yinqiao Powder was established with good separation, and sixteen specific peaks were found. Compared to the decoction of Yinqiao Powder, Yinqiao Jiedu preparations like granules, tablets, pills, oral liquid, and mixture retained 11, 13, 12, 10, 6 specific peaks, respectively, and the dose of chlorogenic acid, forsythoside, arctiin were reduced to various degrees. Conclusion There are much more differences in specific ehromatogram, dose of effective compositions between decoction of Yinqiao Powder and five Yinqiao Jiedu preparations, indicating that more attention shoul be paid to decoction of medicine defects during clinical research and production of patent herbal medicines.
