摘要
目的:探讨信号转导和转录活化因子-3(STAT3)及血管内皮生长因子(VEGF)蛋白在涎腺腺样囊性癌(SACC)组织中的表达情况及其与SACC临床病理参数、血管生成的关系。方法:采用免疫组织化学法检测50例SACC(SACC组)与15例正常腮腺组织(对照组)中STAT3、VEGF蛋白表达和微血管密度(MVD)。结果:(1)SACC组的STAT3及VEGF蛋白表达阳性率分别为66%和72%,明显高于正常组的6.7%和20%(P<0.01)。(2)STAT3和VEGF的表达在SACC的不同组织类型、TNM分期、有无淋巴结转移方面差异均有统计学意义(P<0.05),但在不同性别、年龄及肿瘤部位方面差异均无统计学意义(P>0.05)。(3)STAT3与VEGF表达呈正相关(P<0.05);SACC组的STAT3蛋白及VEGF蛋白表达中阳性与阴性者的MVD差异比较均有统计学意义(均P<0.05)。结论:STAT3可能通过上调VEGF表达,促进SACC中新生血管的形成,STAT3可能会成为SACC的抗血管生成的治疗靶点。
Objective: To investigate expressions of signal transducer and transcription 3 (STAT3) and vascular endothe- lial growth factor (VEGF) in salivary adenoid cystic carcinoma, and explore its relation to elinieopathologieal parameters and tumor angiogenesis. Methods: Expressions of STAT3 and VEGF and values of mierovessel density were detected by immuno- histochemistry in samples of tumor tissues from 50 patients with salivary adenoid cystic carcinoma (SACC group) and those of normal parotid gland tissues from 15 healthy individuals (control group). Results: (1)The positive expression rates of STAT3 and VEGF were 66% and 72% in SACC group, which were significantly higher than those of control group (6.7% and 20%, P 〈 0.01). (2) Expressions of STAT3 and VEGF protein were significantly correlated with SACC clinical pathological type, lymph node metastasis and TNM stage (P 〈 0.05), but no correlation with gender, age and the location of tumor (P 〉 0.05). (3) There was a positive correlation between STAT3 and VEGF in salivary adenoid cystic carcinoma (P 〈 0.05). In 50 specimens of salivary adenoid cystic carcinoma, there was a significant difference in MVD between positive expression and negative ex- pression of STAT3 and VEGF sub-groups in SACC group (P 〈 0.05). Conclusion: By raising VEGF, STAT3 might promote the formation of new blood vessels in SACC. STAT3 pathway can be a new way to prevent angiogenesis of SACC.
