乳香提取物对免疫低下实验小鼠T细胞功能的影响 被引量:2

Boswellic extract enhances T cell function in immunosuppressive mice induced by cyclophosphamide

摘要 研究乳香提取物提高小鼠免疫功能的机理与途径,探索乳香提取物对增强免疫功能的应用潜能。采用环磷酰胺制备免疫低下小鼠模型,以乳香提取物经灌胃给予免疫低下小鼠,连续10d。灌胃结束后取小鼠外周血采用荧光抗体染色并经流式细胞仪检测小鼠T细胞数量变化;取小鼠脾脏单个核细胞加抗CD3和CD28刺激并经3 H-TdR掺入试验,观察小鼠淋巴细胞增殖能力;取小鼠骨髓前体细胞与GM-CSF共培养,3 H-TdR掺入试验观察小鼠骨髓前体细胞增殖能力;ELISA方法检测小鼠血清中细胞因子。结果表明,给免疫低下模型小鼠口服乳香提取物后可有效增加小鼠总T细胞及CD4+T细胞数量:非服用组T细胞为12%±3%,服用组T细胞为28%±4%,两组相比具有显著差异(P<0.01);而CD4+T细胞从5%±2%(非服用组)增加至22%±3%(服用组),两组相比具有显著性统计学差异(P<0.01)。接受乳香提取物处理小鼠的T细胞对抗CD3和CD28刺激的增殖反应明显增高(P<0.05);IFN-γ、TNF-α、IL-12和IL-4水平上调;骨髓粒系细胞受GM-CSF刺激后增殖上调。因此,乳香提取物具有明显上调小鼠T细胞和Th1和Th2细胞数量和功能,其作用可能与刺激骨髓前体细胞增殖有关。 To reveal the mechanism by which Boswellic extract enhances immunity in cyclophosphamide-induced immunodeficiency mice and to explore its therapeutic potential,an cyclophosphamide-induced immunosuppressive mice model was established.Mice were randomly divided into two groups and each group was given Boswellic extract or water orally.After 10 days of administration,all mice were sacrificed and peripheral blood mononuclear cells(PBMC) were analyzed by FACS with specific antibodies.The splenocytes were stimulated with CD3 and CD28 for 72 h,then pulsed with -thymidine,and cultured for additional 16 h.-thymidine,incorporation was counted and used as proliferation ability of T cells.Bone marrow precursor cells(BMPC) were collected and stimulated with GM-CSF for 2 days and their proliferation was measured by thymidine incorporation.Serum cytokines(IL-2,IFN-γ,IL-12,TNF-α,IL-4,IL-17,IL-10 and SCF) were detected by ELISA.The results showed that treatment with Boswellic extract can notably increase the number of T cells and CD4+T cells in peripheral blood,percentage of T and CD4+T cells and T cells proliferation stimulated with CD3 and CD28.Meanwhile,serum levels of IFN-γ,TNF-α and IL-12 in the treated mice was significantly increased.Furthermore,proliferation of BMPC was significantly increased upon GM-CSF stimulation.In summary,Boswellic extract can up-regulate the number and function of Th1 cell obviously.The possible mechanism might be that Boswellic extract induces expansion of BMPC in vivo.
