泄浊解毒方对溃疡性结肠炎大鼠TLR2及IL-8IL-1β影响的研究 被引量:5

The Effects of Xiezhuo Jiedu Decoction on the Expression of TLR2,IL-8and IL-1β in Ulcerative Colitis Rats

摘要 目的:针对溃疡性结肠炎(Ulcerative Colitis,UC)病机组成"泄浊升清、解毒祛瘀"之泄浊解毒方,通过动物实验,观察泄浊解毒方对溃疡性结肠炎大鼠TLR2、IL-8及IL-1β的影响,探讨该方治疗UC可能的作用机制。方法:本研究采用三硝基苯磺酸(2,4,6-trinitrobenzene-sulfonic acid,TNBS)/乙醇联合造模法诱导大鼠溃疡性结肠炎动物模型,实验分为空白组(BG)、模型组(MG)、泄浊解毒方大剂量组(HG)、泄浊解毒方小剂量组(LG)、柳氮磺胺吡啶组(SG)5组。逆转录-聚合酶链反应(RT-PCR)技术检测大鼠结肠组织TLR2mRNA的表达;免疫组化染色(SP法)检测TLR2蛋白的表达;放免法检测大鼠血清IL-8、IL-1β的含量。结果:泄浊解毒方可明显降低TLR2基因、蛋白水平的表达及IL-8、IL-1β的含量。结论:泄浊解毒方对溃疡性结肠炎大鼠有良好的治疗作用,其作用机制可能与降低大鼠TLR2的表达及IL-8、IL-1β的含量有关。 Objective According to the pathogenesis of ulcerative colitis (UC), we composed Xiezhuo Jiedu Decoction, the effect of which was purging turbidity, ascending lucidity, detoxifying and removing blood stasis. We carried out the animal ex- periment to explore the effect of Xiezhuo Jiedu decoction on TLR2, IL-8, IL-1β in ulcerative colitis model rats, and elucidated the possible mechanism of this decoction on ulcerative colitis. Methods: 2, 4, 6-trinitrobenzene-sulfonic acid (TNBS) / etha- nol solution was administrated by enema to make ulcerative colitis rats models. Five groups were divided in the experiments: nor-real control group, modeling control group, Xiezhuo Jiedu Decoction high-dose group, Xiezhuo Jiedu Decoction small-dose group, Salazosulfapyridine group. The expression of TLR2mRNA was detected by RT-PCR; the expression of TLR2protein was dectected by immunohistochemical staining; the contents of IL-8and IL-1β were detected by radioimmunoassy. Results: Xiezhuo Jiedu Decoction could significantly reduce the expression of TLR2, IL-8and IL-1β . Conclusion: Xiezhuo Jiedu Decoction has good therapeutic on decreasing the expression of TLR2mRNA gene and TLR2protein, the content of IL-8and IL-1β are also de-creased. Its mechanism may be related to reduce the expression of TLR2, IL-8and IL-1β .
