摘要 目的研究骨桥蛋白(osteopontin,OPN)和MMP-9在喉鳞状细胞癌(laryngeal squamous cell carcinoma,LSCC)组织中的表达及与临床病理特征之间的相关性,分析两者在LSCC发生、发展过程中的机制。方法采用免疫组化SP法检测47例LSCC及10例正常喉咽黏膜中OPN和MMP-9的表达。结果 (1)47例LSCC组织中OPN、MMP-9蛋白的阳性率分别为63.8%、68.1%,在癌旁正常组织中阳性率为10%和0,两者比较差异有显著性(P<0.05);(2)OPN表达与LSCC临床分期、病理分级、淋巴结转移有关(P<0.05),MMP-9与LSCC临床分期、淋巴结转移有关(P<0.05),OPN和MMP-9的表达与患者年龄、性别、肿瘤位置无关(P>0.05);(3)LSCC组织中OPN和MMP-9的表达呈正相关(r=0.340,P=0.020)。结论 LSCC的发生、发展与OPN和MMP-9的高表达有关,OPN可能通过上调MMP-9的表达,降解细胞外基质,从而促进肿瘤的转移。 Purpose To investigate the expression of OPN and MMP-9 as well as their correlations with the clinical pathological characteristics of laryngeal squamous cell carcinomas(LSCC),and to analyze the potential mechanisms during the development and progression of LSCC.Methods Immunohistochemistry was used to detect the expression of OPN and MMP-9 in 47 cases of LSCC and 10 cases of normal mucosa.Results(1) The expression rates of OPN and MMP-9 in LSCC were 63.8% and 68.1%,respectively,which was significant difference from that in the tumor adjacent issue(10% and 0,respectively).The positive rate of OPN was significantly correlated with the clinical staging,the pathological grading,the metastasis to cervical lymph node(P0.05).(2) The positive rate of MMP-9 was correlated with the clinical staging,the metastasis to cervical lymph node(P0.05),but the positive rates of OPN and MMP-9 were not correlated with patient's age,sex,tumor location(P0.05).(3) The expression of OPN was positively correlated with MMP-9 in LSCC(r=0.340,P=0.020).Conclusion The over-expression of OPN and MMP-9 is correlated with occurrence and development of LSCC,and tumor metastasis might be caused by the enhanced expression of MMP-9,regulated by OPN,leading to the degradation of extracellular matrix.
