摘要
目的研究唐山市汉族退休男性吸烟与纤维蛋白原(Fg)Bβ链位点基因多态性和血浆Fg水平及其聚合功能的关系。方法选取唐山市开滦集团汉族退休男性职工890人作为调查对象,均抽取清晨空腹静脉血测定白细胞介素(IL)、血浆Fg水平和纤维蛋白单体聚合反应速率(FMPV)、最大光密度(Ama)x、FMPV/Amax等反映Fg聚合功能的指标,应用聚合酶链反应-限制性内切酶片段长度多态性技术检测FgBβ链-854G/A、-455G/A、-249C/T、-148C/T、448G/A和Bcl-1G/A6个位点的基因多态性。结果男性吸烟和不吸烟人群均表现为FgBβ-854位点变异基因型者的血浆Fg系列指标高于野生基因型人群,差异有统计学意义(P<0.05)。FgBβ-249位点变异基因型人群中男性吸烟者的血浆Fg水平、FMPV及FMPV/Amax均明显高于不吸烟人群,差异有统计学意义(P<0.05)。结论 FgBβ-854基因多态性位点是影响血浆Fg水平和分子活性功能表达的重要基因位点。吸烟可能与Fg Bβ-249基因型相互作用而使血浆Fg水平升高,并可能影响Bβ-854基因型表达而使血浆Fg分子活性功能增强。
Objective To study the correlation of smoking in Han male retiree of Tangshan with Bβ-854G/A,-455G/A, -249C/T, -148C/T, 448G/A, Bcl-IG/A gene polymorphisms of fibrinogen(Fg), plasma Fg concentration and molecular reactivity. Methods 890 Han male retirees from Tangshan Kailuan Corporation were enrolled. All subjects were phlebotomized in the early morning to measure serum interleukin (IL), Fg concentration, fibrin monomer polymerized velocity (FMPV), absorbance maximum (A max) and FMPV/A max, furthermore, to detect six loci of the gene polymorphisms. The characters of the gene polymorphisms and functional expression of Fg based on smoking status were analyzed. Results In both the groups, the serial indexes of Fg with mutated genotype of Bβ-854 were all higher than those with wild genotypes (P〈0.05); Fg concentration, FMPV and FMPV/A max in smoking group with Bβ-249 mutated genotype were higher than those in non-smoking group (P〈0.05). Conehtsion Fg Bβ-854 is one of main controlling gene loci that influenced plasma Fg concentration and molecular reactivity. Man smoking can increase the Fg concentration through interacting with Bβ-249 mutated genotype, and also affect the expression of Fg conglomerate function by starting effect of Bβ-854 gene locus.
