摘要
以秀丽隐杆线虫为模式生物,建立抗菌物质活体筛选模型.以铜绿假单孢菌感染线虫后,加入不同剂量已知抗生素治疗,观察线虫存活情况及体内细菌数量变化,确定模型建立的操作条件.结果表明,线虫感染程度可量化操作,最佳操作参数为:铜绿假单胞菌培养时间10~12 h,菌液浓度A600值1.0~1.2,线虫感染时间15 h.抗生素的使用能够显著提高感染线虫存活状况,其中环丙沙星治疗浓度范围为0~20μg/mL,线虫存活率和药物剂量之间具有稳定的线性关系.
Caenorhabditis elegans are used to establish screening model for antimicrobial substances. C. elegans are infected with Pseudomonas aeruginosa, and then treated by different doses of known antibiotic. The survival rate of nematodes and the number of in vivo bacterial are observed, the operating conditions of the model establishment are determined. The results show that the operation of nematodes infection degree can be quantified. The optimal oper- ating parameters are as follows : pseudomonas aeruginosa training time 10 - 12 h, bacterium concentration Am 1.0 - 1.2 and infection time 15 h. Antibiotics can significantly prolong the life span of C. elegans. , and the effect of ciprofloxacin treatment indicates that there is stable linear relationship between survival rates of nematodes and antibiotics dose in concentration range of 0 -20 μg/mL.
