摘要
为探讨中医药防治自然流产的机理 ,以利血平、米非司酮 (RU4 86 )建立病证结合动物流产模型 ,模拟脾虚、黄体不健的状态 ,观察补肾健脾中药复方“助孕 3号方”对该模型流产率的影响 ;并以四君子汤、黄体酮为对照 ,同时设立脾虚流产模型组、脾虚模型组、流产模型组及正常妊娠组 ;以血清孕激素水平、子宫蜕膜孕激素受体 (PR)阳性率及子宫卵巢病理形态学变化为观察指标。结果 :助孕 3号组流产率低于四君子汤组、流产模型组 (P <0 .0 5) ;该组孕激素水平、PR阳性率均比四君子汤组、脾虚流产模型组、流产模型组高 ;并对脾虚大鼠体重的减轻有改善作用。提示 :“助孕 3号方”是通过促进妊娠黄体和合体滋养细胞的分泌功能 ,维持靶细胞受体的含量及改善脾虚证候、提高整体功能而发挥效应。说明其作用是多环节、多层次的。同时表明脾肾并补法优于单纯健脾法 。
To explore the mechanism of Zhuyun Ⅲ, a herbal formula for spontaneous abortion by strengthening the spleen and supplementing the kidney, an abortion model with corpus luteum inhibition induced by reserpine and mifepristone (RU486) was used. One_hundred and four pregnant SD rats were allocated to seven groups:①reserpine+RU486+Zhuyun Ⅲ, ②reserpine+RU486+sijunzitang, ③reserpine+RU486+progesterone,④reserpine+RU486,⑤reserpine control,⑥RU486 control and ⑦ normal pregnant group. Different groups were observed at the same period on the level of serum progesterone(P),estradio(E 2), follicle_stimulating hormone(FSH), the positive rate of decidua progesterone receptor(PR) and the pathological changes of uterus and ovaries. The results showed that the abortion rate of group ① was lower than that of group ④ and ⑥(P<0.05). The level of serum progesterone and the positive rate of PR of group ① were all higher than that of group ②, ④ and ⑥. On the other hand, Zhuyun Ⅲ can also improve the weight reduction of body, spleen and ovary. The results indicate that Zhuyun Ⅲ exerts anti_abortion effect by promoting the secretion of corpus luteum and trophocytes, maintaining the content of target cells' receptor and relieving spleen_deficiency condition. It is also suggested that strengthening the spleen and kidney is better than supplementing the spleen only.
