摘要
[目的]建立高效液相色谱法测定半夏中β-谷甾醇的含量。[方法]色谱条件:色谱柱为岛津Shim-Pack VP-ODS C18(4.6 mm×150 mm,5μm),流动相为无水甲醇,流速为1.0 ml/min,检测波长为210 nm,柱温为35℃。[结果]β-谷甾醇在0.200~2.000μg/ml浓度范围内线性关系良好,r=0.999 6,平均回收率为101.07%(n=5),RSD为2.96%;半夏样品中β-谷甾醇含量的平均含量为1.59mg/g。[结论]该方法分离度较好,可用于半夏药材中β-谷甾醇的含量测定。
[Objective] The aim was to establish a HPLC method for determination of β-sitosterol in Pinellia ternata(Thunb.)Breit.[Method] The Shim-Pack VP-ODS C18 column(4.6 mm×150 mm 5 μm)was used,and mobile phase was composed of CH3OH.Detection wavelength was at 210 nm,the flow rate was 1.0 ml/min,and column temperature was at 35 ℃.[Result] The linear response range from 0.206-2.060 μg/ml,r=0.999 6.The recovery rate was 101.07%(n=5),RSD was 2.96%.[Conclusion] The HPLC method was accurate and suitable for the determination of β-sitosterol in Pinellia ternata(Thunb.)Breit..
