摘要
[目的]研究白术人工种子最佳胚乳配方,为白术人工种子技术规模化开发应用提供理论依据。[方法]以白术试管苗无菌体系为试验材料,研究了人工种子包埋繁殖体、人工胚乳配方对人工种子萌发率和成苗率的影响。[结果]以腋芽为包埋繁殖体,2.5%海藻酸钠+2%CaCl2为人工胚乳基质,0.5 mg/L IAA和30 g/L蔗糖为人工胚乳配方,播种后萌发率和成苗率分别为100%和92%。[结论]以组织培养为基础的白术人工种子快速繁殖技术具有一定的优越性。
[Objective] To study the best synthetic endosperm combination of Atractylodes macrocephala Koidz.Seeds and to provide the theoretical basis for development and large-scale application of A.macrocephala synthetic seeds.[Method] Factors to affect germination rate and seedling conversion rate,such as encapsulated propagule and synthetic endosperm components were studied with germfree tube plantlet system of A.macrocephala.[Result] The germination rate and seedling conversion rate of synthetic seeds could reach 100% and 92% with auxilizry buds,2.5% sodium alginate + 2% CaCl2 as synthetic endosperm encapsulation,and 0.5 mg/L IAA and 30 g/L sucrose as synthetic endosperm components.[Conclusion] The technique of rapid multiplication of synthetic seeds on the basis of tissue culture of A.macrocephala has definite superiority.
